Alginate-based nanomaterials: Fabrication techniques, properties, and applications

Polysaccharides, such as cellulose, hyaluronic acid, alginic acid, and chitosan, as well as polysaccharide derivatives, have been successfully used to augment drug delivery in the treatment of ocular pathologies. The properties of polysaccharides can be extensively modified to optimize ocular drug formulations and to obtain biocompatible and biodegradable drugs with improved bioavailability and tailored pharmacological effects. This review discusses the available polysaccharide choices for overcoming the difficulties associated with ocular drug delivery, and it explores the reasons for the dependence between the physicochemical properties of polysaccharide-based drug carriers and their efficiency in different formulations and applications. Polysaccharides will continue to be of great interest to researchers endeavoring to develop ophthalmic drugs with improved effectiveness and safety.

Polysaccharides in Ocular Drug Delivery

The invention relates to a method of nanofibres production from a polymer solution using electrostatic spinning in an electric field created by a potential difference between a charged electrode and a counter electrode. The polymer solution (2) is for spinning supplied into the electric field using the surface of the rotating charged electrode (30), while on a part of the circumference of the charged electrode (30) near to the counter electrode (40) is a spinning surface created, by which is a high spinning capacity reached. Further the invention relates to a device for carrying out the method, where the charged electrode (30) is pivoted and by its (bottom) part of its circumference it is immersed in the polymer solution (2), while against the free part of the circumference of the charged electrode (30) is positioned the counter electrode (40).

Pueraria tuberosa is known for its therapeutic potentials in cardiovascular disorders, but its effect in angiogenesis has not been studied so far. In this study, a computational approach has been applied to elucidate the role of the phytochemicals in inhibition of angiogenesis through modulation of vascular endothelial growth factor receptors: Vascular endothelial growth factor receptor-1 and vascular endothelial growth factor receptor-2, major factors responsible for angiogenesis. Metabolite structures retrieved from PubChem and KNApSAcK - 3D databases, were docked using AutoDock4.2 tool. Hydrogen bond and molecular docking, absorption, distribution, metabolism and excretion and toxicity predictions were carried out using UCSF Chimera, LigPlot(+) and PreADMET server, respectively. From the docking analysis, it was observed that puerarone and tuberostan had significant binding affinity for the intracellular kinase domain of vascular endothelial growth factor receptors-1 and vascular endothelial growth factor receptor-2 respectively. It is important to mention that both the phytochemicals shared similar interaction profile as that of standard inhibitors of vascular endothelial growth factor receptors. Also, both puerarone and tuberostan interacted with Lys861/Lys868 (adenosine 5'-triphosphate binding site of vascular endothelial growth factor receptors-1/vascular endothelial growth factor receptors-2), thus providing a clue that they may enforce their inhibitory effect by blocking the adenosine 5'-triphosphate binding domain of vascular endothelial growth factor receptors. Moreover, these molecules exhibited good drug-likeness, absorption, distribution, metabolism and excretion properties without any carcinogenic and toxic effects. The interaction pattern of the puerarone and tuberostan may provide a hint for a novel drug design for vascular endothelial growth factor tyrosine kinase receptors with better specificity to treat angiogenic disorders.

Molecular Docking and Interactions of Pueraria Tuberosa with Vascular Endothelial Growth Factor Receptors.

The present study describes the preparation of extracellular matrix (ECM; from porcine omentum) based chitosan composite films for wound dressing applications. The films were prepared by varying the ECM content, whereas, the amount of chitosan was kept constant. The interactions amongst the components of the films were analyzed by FTIR and XRD studies. The films were thoroughly characterized for surface hydrophilicity, moisture retention capability, water vapor permeability, mechanical and biocompatibility. FTIR study indicated that both chitosan and ECM were present in their native form and did not lose their activity. XRD analysis suggested composition dependent change in the crystallinity of the films. The mechanical properties suggested that the composite films had sufficient properties to be used for wound dressing applications. An increase in the ECM content resulted in better hydrophilicity of the films and hence better the moisture retention capacity and retardant water vapor transmission rate property of the composite films. The films were found to be biocompatible to both blood and adipose tissue derived stem cells. In gist, the prepared films may be explored as wound dressing materials.

Evaluation extracellular matrix-chitosan composite films for wound healing application

The current study describes the synthesis and characterization of dual environment-responsive semi-interpenetrating hydrogels of polyhydroxyethyl methacrylate (pHEMA) and methyl cellulose (MC). HEMA was polymerized using free radical polymerization technique. Different proportions of MC were incorporated within the pHEMA matrix to induce environment-sensitive property. The microstructures of the hydrogels were studied under bright field microscopy. The hydrogels were thoroughly characterized using XRD, FTIR, swelling, mechanical and electrical studies. The micrographs of the hydrogels suggested formation of biphasic system. At lower proportions of MC, the hydrogels were oil-in-water type of formulations. An increase in the MC content resulted in the formation of bicontinuous biphasic formulations. Mechanical analysis suggested viscoelastic solid nature of the hydrogels. Synthesis and characterization of novel dual environment-responsive hydrogels of Hydroxyethyl methacrylate and Methyl cellulose

The study describes the optimization of the process parameters for the fabrication of nano- fibrous scaffolds of poly (vinyl alcohol) (PVA) and sodium alginate (SA) blends using nozzle free electrospinning machine. The solutions prepared by varying the proportions of PVA and SA solutions were characterized by surface tensiometer and conductivity meter which showed reduction in the surface tension and conductivity of the solutions with increase in PVA concentration. The process optimization was tried by tailoring the applied voltage and roller - collector distance to obtain maximum production with least diameter fibers. The scaffolds were characterized by SEM, FT-IR, XRD and TG studies. The applied voltage, roller-collector distance of the optimized scaffold was 75 kV, 12cm respectively. A decrease in fiber diameter was observed with the increase in the proportion of SA. Inter - molecular hydrogen bonding between PVA and SA molecules and the amorphous nature of the scaffolds was suggested through FTIR and XRD studies respectively.
