Journal ArticleDOI
ATP release and purinergic signaling: a common pathway for particle-mediated inflammasome activation.
Nicolas Riteau,Ludivine Baron,B. Villeret,Noëlline Guillou,F. Savigny,Bernhard Ryffel,François Rassendren,M. Le Bert,Aurélie Gombault,Isabelle Couillin +9 more
TL;DR: It is shown that uric acid, silica and Alum crystals trigger the extracellular delivery of endogenous ATP, which just precedes the secretion of mature interleukin-1β by macrophages, both events depending on purinergic receptors and connexin/pannexin channels.
Journal ArticleDOI
The NLRP3 inflammasome is activated by nanoparticles through ATP, ADP and adenosine
Ludivine Baron,Aurélie Gombault,Manoussa Fanny,B. Villeret,F. Savigny,Noëlline Guillou,Corinne Panek,M. Le Bert,Vincent Lagente,François Rassendren,Nicolas Riteau,Isabelle Couillin +11 more
TL;DR: It is shown for the first time that extracellularAdenosine activates the NLRP3 inflammasome by two ways: by interacting with adenosine receptors at nanomolar/micromolar concentrations and through cellular uptake by equilibrative nucleoside transporters at millimolar concentrations.
Journal ArticleDOI
B cell activating factor is central to bleomycin- and IL-17-mediated experimental pulmonary fibrosis
Antoine François,Aurélie Gombault,B. Villeret,Ghada Alsaleh,Manoussa Fanny,Paméla Gasse,Sylvain Marchand Adam,Bruno Crestani,Jean Sibilia,Pascal Schneider,Seiamak Bahram,Valérie F. J. Quesniaux,Bernhard Ryffel,Dominique Wachsmann,Jacques-Eric Gottenberg,Isabelle Couillin +15 more
TL;DR: A role for BAFF in the establishment of pulmonary fibrosis and a crosstalk between IL-1β, BAFF and IL-17A is supported and elevated levels of BAFF are reported in bronchoalveolar lavages from IPF patients.
Journal ArticleDOI
Discordance in cathepsin B and cystatin C expressions in bronchoalveolar fluids between murine bleomycin-induced fibrosis and human idiopathic fibrosis
Mariana Kasabova,Mariana Kasabova,B. Villeret,Aurélie Gombault,Fabien Lecaille,Thomas Reinheckel,Sylvain Marchand-Adam,Isabelle Couillin,Gilles Lalmanach +8 more
TL;DR: The activity of cysteine cathepsin B increased markedly in lung homogenates and in bronchoalveolar lavage fluids (BALF) of the mouse model of bleomycin-induced lung fibrosis after 14 days of challenge, and cystatin C was unaffected in BALF of wild-type and cathePSin B-deficient mice.
Journal ArticleDOI
B-cell activating factor regulates IL-1β- and IL-17A-mediated pulmonary fibrosis in mice
Manoussa Fanny,Aurélie Gombault,F. Francois,B. Villeret,Bernhard Ryffel,D. Wachhsmann,Jacques-Eric Gottenberg,Isabelle Couillin +7 more
TL;DR: The present data, demonstrating a critical role of BAFF by amplifying the IL-17A effect in experimental lung fibrosis, together with increased BAFF levels in the airways of patients with IPF, suggest that BAFF might be a therapeutic target of IPF.
