Control of PKR Protein Kinase by Hepatitis C Virus Nonstructural 5A Protein: Molecular Mechanisms of Kinase Regulation
Michael Gale,Collin M. Blakely,Bart Kwieciszewski,Seng Lai Tan,Michelle L. Dossett,Norina M. Tang,Marcus J. Korth,Stephen J. Polyak,David R. Gretch,Michael G. Katze +9 more
TL;DR: Results indicate that mutations within the PKR-binding region of NS5A, including those within the ISDR, can disrupt the NS5a-PKR interaction, possibly rendering HCV sensitive to the antiviral effects of interferon.
Journal ArticleDOI
Antiapoptotic and Oncogenic Potentials of Hepatitis C Virus Are Linked to Interferon Resistance by Viral Repression of the PKR Protein Kinase
TL;DR: Disruption of apoptosis and tumorogenesis required the PKR-binding function of NS5A, demonstrating that these properties may be linked to the IFN-resistant phenotype of HCV.
Journal ArticleDOI
Ser2194 Is a Highly Conserved Major Phosphorylation Site of the Hepatitis C Virus Nonstructural Protein NS5A
Michael G. Katze,Bart Kwieciszewski,David R. Goodlett,Collin M. Blakely,Petra Neddermann,Seng Lai Tan,Rudolph Aebersold +6 more
TL;DR: Two-dimensional phosphopeptide mapping showed that a protein kinase or kinases present in yeast, insect, and mammalian cells phosphorylated a highly purified HCV genotype 1b NS5A from insect cells on identical serine residues by using negative-ion electrospray ionization-microcapillary high performance liquid chromatography-mass spectrometry.
