The Huisgen 1,3-dipolar cycloaddition reaction of organic azides and alkynes has gained considerable attention in recent years due to the introduction in 2001 of Cu(1) catalysis by Tornoe and Meldal, leading to a major improvement in both rate and regioselectivity of the reaction, as realized independently by the Meldal and the Sharpless laboratories. The great success of the Cu(1) catalyzed reaction is rooted in the fact that it is a virtually quantitative, very robust, insensitive, general, and orthogonal ligation reaction, suitable for even biomolecular ligation and in vivo tagging or as a polymerization reaction for synthesis of long linear polymers. The triazole formed is essentially chemically inert to reactive conditions, e.g. oxidation, reduction, and hydrolysis, and has an intermediate polarity with a dipolar moment of ∼5 D. The basis for the unique properties and rate enhancement for triazole formation under Cu(1) catalysis should be found in the high ∆G of the reaction in combination with the low character of polarity of the dipole of the noncatalyzed thermal reaction, which leads to a considerable activation barrier. In order to understand the reaction in detail, it therefore seems important to spend a moment to consider the structural and mechanistic aspects of the catalysis. The reaction is quite insensitive to reaction conditions as long as Cu(1) is present and may be performed in an aqueous or organic environment both in solution and on solid support.

Efficiency and Fidelity in a Click-Chemistry Route to Triazole Dendrimers by the Copper(I)-Catalyzed Ligation of Azides and Alkynes

“Life is branched” was the motto of a special issue of Macromolecular Chemistry and Physics1 on “Branched Polymers”, indicating that branching is of similar importance in the world of synthetic macromolecules as it is in nature. The significance of branched macromolecules has evolved over the last 30 years from just being considered as a side reaction in polymerization or as a precursor step in the formation of networks. Important to this change in perception of branching was the concept of “polymer architectures”, which formed on new starand graft-branched structures in the 1980s and then in the early 1990s on dendrimers and dendritic polymers. Today, clearly, controlled branching is considered to be a major aspect in the design of macromolecules and functional material. Hyperbranched (hb) polymers are a special type of dendritic polymers and have as a common feature a very high branching density with the potential of branching in each repeating unit. They are usually prepared in a one-pot synthesis, which limits the control on molar mass and branching accuracy and leads to “heterogeneous” products with a distribution in molar mass and branching. This distinguishes hyperbranched polymers from perfectly branched and monodisperse dendrimers. In the last 20 years, both classes of dendritic polymers, dendrimers as well as hb polymers, have attracted major attention because of their interesting properties resulting from the branched architecture as well as the high number of functional groups.2 The challenging synthesis of the dendrimers attracted especially scientists with a strong organic chemistry background and led to beautifully designed macromolecules, which allowed a deeper insight into the effect of branching and functionality. Dendrimers have been considered as perfect “nano-objects” where one can control perfectly size and functionality, which is of high interest in nanotechnology and biomedicine. hb polymers, however, were considered from the beginning as products suitable for larger-scale application in typical polymer fields like coatings and resins, where a perfect structure is sacrificed for an easy and affordable synthetic route. Thus, the first structures that were reported paralleled the chemistry used for linear polymers like typical polycondensation for polyester synthesis. More recently, unconventional synthetic methods have been adopted also for hb polymers and related structures. Presently, a vast variety of highly branched structures have been realized and studied regarding their properties and potential application fields. In the last 12 years the field of hyperbranched polymers has been well established with a large variety of synthetic approaches and fundamental studies on structure and properties of these unique materials. However, new developments involving hyperbranched materials appeared recently, for example, different synthetic strategies, new reaction mechanisms, formation of more complex architectures, a deeper understanding of the branched structure and their kinetic development, and intensive studies on the material properties and possible applications. This demonstrates the high versatility and the possibilities that are still involved in hyperbranched polymers and render it one of the most active fields in polymer science with a very promising future. © 2000 John Wiley & Sons, Inc. New developments in hyperbranched polymers

The contamination of aquatic ecosystems with microplastics has recently been reported through many studies, and negative impacts on the aquatic biota have been described. For the chemical identification of microplastics, mainly Fourier transform infrared (FTIR) and Raman spectroscopy are used. But up to now, a critical comparison and validation of both spectroscopic methods with respect to microplastics analysis is missing. To close this knowledge gap, we investigated environmental samples by both Raman and FTIR spectroscopy. Firstly, particles and fibres >500 μm extracted from beach sediment samples were analysed by Raman and FTIR microspectroscopic single measurements. Our results illustrate that both methods are in principle suitable to identify microplastics from the environment. However, in some cases, especially for coloured particles, a combination of both spectroscopic methods is necessary for a complete and reliable characterisation of the chemical composition. Secondly, a marine sample containing particles <400 μm was investigated by Raman imaging and FTIR transmission imaging. The results were compared regarding number, size and type of detectable microplastics as well as spectra quality, measurement time and handling. We show that FTIR imaging leads to significant underestimation (about 35 %) of microplastics compared to Raman imaging, especially in the size range <20 μm. However, the measurement time of Raman imaging is considerably higher compared to FTIR imaging. In summary, we propose a further size division within the smaller microplastics fraction into 500–50 μm (rapid and reliable analysis by FTIR imaging) and into 50–1 μm (detailed and more time-consuming analysis by Raman imaging).

Analysis of environmental microplastics by vibrational microspectroscopy: FTIR, Raman or both?

Fifteen years of research on hyperbranched polymers in the group of Brigitte Voit are described, with a focus first on hyperbranched polyester synthesis and then on the addition and cycloaddition reactions used for the preparation of the hyperbranched structure. The characterization of structural details and bulk, solution, and thin-film properties is highlighted, and steps toward the elucidation of a general property profile of hyperbranched polymers are discussed. Some effects of hyperbranched polymers in reactive formulations and blends and in thin films are addressed that can lead to applications in coatings, as additives, and in microelectronics or sensorics. The great progress possible in the last years is shown, but open questions and unsolved problems are also pointed out. © 2005 Wiley Periodicals, Inc.
