2-hydroxyglutarate detection by magnetic resonance spectroscopy in IDH-mutated patients with gliomas.
Changho Choi,Sandeep K. Ganji,Ralph J. DeBerardinis,Kimmo J. Hatanpaa,Dinesh Rakheja,Dinesh Rakheja,Zoltan Kovacs,Xiao Li Yang,Tomoyuki Mashimo,Jack M Raisanen,Isaac Marin-Valencia,Juan M. Pascual,Christopher J. Madden,Bruce E. Mickey,Craig R. Malloy,Robert Bachoo,Elizabeth A. Maher +16 more
TL;DR: In this article, a non-invasive detection of 2-hydroxyglutarate (2HG) by proton magnetic resonance spectroscopy (MRS) was reported.
Journal ArticleDOI
Acetate Is a Bioenergetic Substrate for Human Glioblastoma and Brain Metastases
Tomoyuki Mashimo,Kumar Pichumani,Vamsidhara Vemireddy,Kimmo J. Hatanpaa,Dinesh K. Singh,Shyam Sirasanagandla,Suraj Nannepaga,Sara Grazia Maria Piccirillo,Zoltan Kovacs,Chan Foong,Zhiguang Huang,Samuel L. Barnett,Bruce E. Mickey,Ralph J. DeBerardinis,Benjamin P. Tu,Elizabeth A. Maher,Robert Bachoo +16 more
TL;DR: The data demonstrate a strikingly common metabolic phenotype in diverse brain tumors that includes the ability to oxidize acetate in the citric acid cycle, which may be important for meeting the high biosynthetic and bioenergetic demands of malignant growth.
Journal ArticleDOI
Analysis of tumor metabolism reveals mitochondrial glucose oxidation in genetically diverse human glioblastomas in the mouse brain in vivo.
Isaac Marin-Valencia,Chendong Yang,Tomoyuki Mashimo,Steve K. Cho,Hyeon-Man Baek,Xiao Li Yang,Kartik N. Rajagopalan,Melissa Maddie,Vamsidhara Vemireddy,Zhenze Zhao,Ling Cai,Levi B. Good,Benjamin P. Tu,Kimmo J. Hatanpaa,Bruce E. Mickey,José M. Matés,Juan M. Pascual,Elizabeth A. Maher,Craig R. Malloy,Craig R. Malloy,Ralph J. DeBerardinis,Robert Bachoo +21 more
TL;DR: GBM cells utilize mitochondrial glucose oxidation during aggressive tumor growth in vivo, and many of these same activities were conserved in cells cultured ex vivo from the tumors.
Journal ArticleDOI
Metabolism of [U‐13C]glucose in human brain tumors in vivo
Elizabeth A. Maher,Isaac Marin-Valencia,Robert Bachoo,Tomoyuki Mashimo,Jack M Raisanen,Kimmo J. Hatanpaa,Ashish Jindal,F. Mark H Jeffrey,Changho Choi,Christopher J. Madden,Dana Mathews,Juan M. Pascual,Bruce E. Mickey,Craig R. Malloy,Craig R. Malloy,Ralph J. DeBerardinis +15 more
TL;DR: A convenient approach is illustrated that capitalizes on the high information content of 13C NMR spectroscopy and enables the analysis of intermediary metabolism in diverse cancers growing in their native microenvironment.
Journal ArticleDOI
EGFRvIII and DNA Double-Strand Break Repair: A Molecular Mechanism for Radioresistance in Glioblastoma
Bipasha Mukherjee,Brian McEllin,Cristel V. Camacho,Nozomi Tomimatsu,Shyam Sirasanagandala,Suraj Nannepaga,Kimmo J. Hatanpaa,Bruce E. Mickey,Christopher J. Madden,Elizabeth A. Maher,David A. Boothman,Frank B. Furnari,Webster K. Cavenee,Robert M. Bachoo,Sandeep Burma +14 more
TL;DR: In vitro and in vivo data support the hypothesis that EGFRvIII expression promotes DNA-PKcs activation and DSB repair, perhaps as a consequence of hyperactivated PI3K/Akt-1 signaling, and raise the possibility that EG FR and/or DNA- PKcs inhibition concurrent with radiation may be an effective therapeutic strategy for radiosensitizing high-grade gliomas.
