Opa1 Mutations Induce Mitochondrial Dna Instability and Optic Atrophy Plus Phenotypes
Valerio Carelli,Pascal Reynier,Maria Lucia Valentino,Rosanna Carroccia,Luisa Iommarini,C. La Morgia,Rafael Garesse,Guy Lenaers,Dominique Bonneau,Patrizia Amati-Bonneau +9 more
TL;DR: In this paper, mutations in the OPA1 gene can also be responsible for a syndromic form of DOA associated with sensorineural deafness, ataxia, axonal sensory-motor polyneuropathy, chronic progressive external ophthalmoplegia and mitochondrial myopathy.
Journal ArticleDOI
Cigarette toxicity triggers Leber's hereditary optic neuropathy by affecting mtDNA copy number, oxidative phosphorylation and ROS detoxification pathways.
Luca Giordano,Stefania Deceglie,Pio D'Adamo,Maria Lucia Valentino,C. La Morgia,Flavio Fracasso,Marina Roberti,M Cappellari,Giuseppe Petrosillo,S Ciaravolo,Dino Parente,Carla Giordano,Alessandra Maresca,Luisa Iommarini,V. Del Dotto,Anna Ghelli,Solange Rios Salomão,Adriana Berezovsky,Rubens N. Belfort,Alfredo A. Sadun,Valerio Carelli,P. Loguercio Polosa,Palmiro Cantatore,Palmiro Cantatore +23 more
TL;DR: Exposure to smoke derivatives has a more deleterious effect in affected individuals, whereas carriers are the most efficient in mitigating ROS rather than recovering bioenergetics, according to carriers' success in reactive oxygen species (ROS) scavenging capacity.
Journal ArticleDOI
Loss of temporal retinal nerve fibers in Parkinson disease: a mitochondrial pattern?
C. La Morgia,Piero Barboni,Giovanni Rizzo,Michele Carbonelli,Giacomo Savini,Cesa Scaglione,Sabina Capellari,Sara Bonazza,Maria Pia Giannoccaro,Giovanna Calandra-Buonaura,Rocco Liguori,Pietro Cortelli,Paolo Martinelli,Agostino Baruzzi,Valerio Carelli +14 more
TL;DR: This data indicates that optic nerve involvement is frequent in mitochondrial disease, and retinal abnormalities are described in Parkinson’s disease.
Journal ArticleDOI
Rare mtDNA variants in Leber hereditary optic neuropathy families with recurrence of myoclonus
C. La Morgia,Alessandro Achilli,Luisa Iommarini,Piero Barboni,Maria Pala,Anna Olivieri,Claudia Zanna,Sara Vidoni,Caterina Tonon,Raffaele Lodi,Roberto Vetrugno,Barbara Mostacci,Rocco Liguori,Rosanna Carroccia,Pasquale Montagna,Michela Rugolo,Antonio Torroni,Valerio Carelli +17 more
TL;DR: The sequence analysis suggests that the LHON plus phenotype in cases may relate to the synergic role of mtDNA variants, which are common to both families.
Journal ArticleDOI
Diffusion Tensor Imaging Mapping of Brain White Matter Pathology in Mitochondrial Optic Neuropathies
David Neil Manners,Giovanni Rizzo,C. La Morgia,Caterina Tonon,Claudia Testa,Piero Barboni,Emil Malucelli,Maria Lucia Valentino,Leonardo Caporali,D. Strobbe,Valerio Carelli,Raffaele Lodi +11 more
TL;DR: Patients with Leber hereditary optic neuropathy had preferential involvement of the optic and acoustic radiations, consistent with trans-synaptic degeneration, whereas patients with optic atrophy gene 1-autosomal dominant opticatrophy presented with widespread involvement suggestive of a multisystemic, possibly a congenital/developmental, disorder.
