Nature relies on large molecules to carry out sophisticated chemical operations, such as catalysis, tight and specific binding, directed flow of electrons, or controlled crystallization of inorganic phases. The polymers entrusted with these crucial tasks, mostly proteins but sometimes RNA, are unique relative to other biological and synthetic polymers in that they adopt specific compact conformations that are thermodynamically and kinetically stable. These folding patterns generate “active sites” via precise three-dimensional arrangement of functional groups. In terms of covalent connectivity, the groups that comprise the active site are often widely spaced along the polymer backbone. The remarkable range of chemical capabilities that evolution has elicited from proteins suggests that it might be possible to design analogous capabilities into unnatural polymers that fold into compact and specific conformations. Since biological evolution has operated under many constraints, the functional properties of proteins and RNA should be viewed as merely exemplifying the potential of compactly folded polymers. The chemist’s domain includes all possible combinations of the elements, and the biological realm, vast and complex though it may be, is only a small part of that domain. Therefore, realization of the potential of folding polymers may be limited more by the human imagination than by physical barriers. I use the term “foldamer” to describe any polymer with a strong tendency to adopt a specific compact conformation. Among proteins, the term “compact” is associated with tertiary structure, and there is as yet no synthetic polymer that displays a specific tertiary structure. Protein tertiary structure arises from the assembly of elements of regular secondary structure (helices, sheets, and turns). The first step in foldamer design must therefore be to identify new backbones with well-defined secondary structural preferences. “Well-defined” in this case means that the conformational preference should be displayed in solution by oligomers of modest length, and I will designate as a foldamer any oligomer that meets this criterion. Within the past decade, a handful of research groups have described unnatural oligomers with interesting conformational propensities. The motivations behind such efforts are varied, but these studies suggest a collective, emerging realization that control over oligomer and polymer folding could lead to new types of molecules with useful properties. The purpose of this “manifesto” is to introduce a large audience to the broad research horizons offered by the concept of synthetic foldamers. The path to creating useful foldamers involves several daunting steps. (i) One must identify new polymeric backbones with suitable folding propensities. This goal includes developing a predictively useful understanding of the relationship between the repetitive features of monomer structure and conformational properties at the polymer level. (ii) One must endow the resulting foldamers with interesting chemical functions, by design, by randomization and screening (“evolution”), or by some combination of these two approaches. (iii) For technological utility, one must be able to produce a foldamer efficiently, which will generally include preparation of the constituent monomers in stereochemically pure form and optimization of heteropolymer synthesis. Each of these steps involves fascinating chemical challenges; the first step is the focus of this Account.

A field guide to foldamers.

A new iron(III) halide-promoted aza-Prins cyclization between γ,δ-unsaturated tosylamines and aldehydes provides six-membered azacycles in good to excellent yields. The process is based on the consecutive generation of γ-unsaturated-iminium ion and further nucleophilic attack by the unsaturated carbon−carbon bond. Homoallyl tosylamine leads to trans-2-alkyl-4-halo-1-tosylpiperidine as the major isomer. In addition, the alkyne aza-Prins cyclization between homopropargyl tosylamine and aldehydes gives 2-alkyl-4-halo-1-tosyl-1,2,5,6-tetrahydropyridines as the only cyclic products. The piperidine ring is widely distributed throughout Nature, e.g., in alkaloids,1 and is an important scaffold for drug discovery, being the core of many pharmaceutically significant compounds.2,3 The syntheses of these type of compounds have been extensively studied in the development of new drugs containing six-membered-ring heterocycles.4 Reactions between N-acyliminium ions and nucleophiles, also described as amidoalkylation or Mannich-type condensations, have been frequently used to introduce substituents at the R-carbon of an amine.5 There are several examples that involve an intramolecular attack of a nucleophilic olefin into an iminium cation for the construction of a heterocyclic ring system.6 Traditionally, the use of hemiaminals or their derivatives as precursors of N-acyliminium intermediates has been a common two-step strategy in these reactions.6a Among this type of cyclization is the aza-Prins cyclization,7 which uses alkenes as intramolecular nucleophile. Schiff base ligands are considered “privileged ligands” because they are easily prepared by the condensation between aldehydes and imines. Stereogenic centres or other elements of chirality (planes, axes) can be introduced in the synthetic design. Schiff base ligands are able to coordinate many different metals, and to stabilize them in various oxidation states, enabling the use of Schiff base metal complexes for a large variety of useful catalytic transformations. Practical guidelines for the preparation and use of different Schiff base metal complexes in the field of catalytic transformations are discussed in this tutorial review.

The discovery of efficient methods for gaining access to enantiomerically pure compounds in the development of pharmaceuticals, agrochemicals, and flavors has been a substantial challenge for chemists. Among the various ways to produce enantiopure compounds, enantioselective homogeneous catalysis constitutes a very appealing strategy as witnessed by the large number of publications in this field and the award of the Nobel Prize 2001 to Knowles,1 Noyori,2 and Sharpless.3 In this approach, a substoichiometric amount (down to 10-3 mol %) of a chiral substance (usually a transition-metal complex obtained from a chiral ligand but also with increasing interest a purely organic molecule) catalyzes the transformation of a prochiral substrate while at the same time dissymmetrically shaping the space around the reaction center, so that one stereochemical path is preferentially followed. The choice of an appropriate chiral ligand is perhaps most crucial, its structure being often the result of knowledge-based intuition or serendipity. In addition, an efficient enantioselective catalytic system is usually obtained after extensive trial-and-error optimization of a number of concurrent factors (ligand structure, metal ion, stoichiometry, solvent, temperature, etc.). In such a complex scenario, combinatorial methodologies may be seen as a response to the requirement of new, more active, and selective catalysts for organic transformations of interest. The application of combinatorial chemistry to enantioselective homogeneous catalysis has lagged behind its use in other areas (e.g., pharmaceuticals lead discovery and optimization). However, in the past seven years there has been accelerated progress as witnessed by the increase of publications in this field (only 2 in 1995 and about 40 in 2002) and the several recent reviews which have covered this subject (Gennari,4 Hoveyda,5,6 Jandeleit,7,8 Bräse,9 Reetz,10,11 Gilbertson,12 de Vries13). An additional important consideration in favor of the creation of libraries of chiral catalysts is in regard to the catalyst scope and applicability. In fact, enzymes are typically substrate-specific, whereas synthetic catalysts usually possess a broader substrate scope. However, there is no general synthetic catalyst, one that is good for every reaction and every substrate. Diversity is important and necessary: a flexible generation of catalyst libraries enhances the chances to find systems with optimum performance (reactivity and selectivity) for different substrates.

Synthesis of Sulfonamido-Pseudopeptides: New Chiral Unnatural Oligomers

The use of non covalent supramolecular ligand–ligand and ligand–substrate interactions in transition metal-catalysed transformations is a new, rapidly emerging area of research. Non-covalent interactions between monodentate ligands such as hydrogen bonding, coordinative bonding, ion pairing, π–π interactions and the formation of inclusion compounds, have been shown to impart higher activity and chemo-, regio-, and stereoselectivity to the corresponding transition metal complexes in a number of catalytic applications. Analogously, supramolecular ligand–substrate interactions, and particularly hydrogen bonding, have been used to direct the regio- and stereochemistry of several metal-catalysed reactions. The catalytic systems relying on supramolecular interactions are generally capable of self-assembling from simpler components in the environment where catalysis is to take place, and are therefore very well-suited for combinatorial catalyst discovery strategies and high-throughput screening.
