Design strategies for water-soluble small molecular chromogenic and fluorogenic probes.

Due to the biological importances of thiols, such as cysteine, homocysteine and glutathione, the development of optical probes for thiols has been an active research area in recent few years. This critical review focuses on the fluorescent or colorimetric sensors for thiols according to their unique mechanisms between sensors and thiols, including Michael addition, cyclization with aldehyde, cleavage of sulfonamide and sulfonate ester by thiols, cleavage of selenium–nitrogen bond by thiols, cleavage of disulfide by thiols, metal complexes-oxidation–reduction, metal complexes-displace coordination, nano-particles and others (110 references).

Fluorescent and colorimetric probes for detection of thiols

In this perspective we introduce the basic photophysics of the excited-state intramolecular proton transfer (ESIPT) chromophores, then the state-of-the-art development of the ESIPT chromophores and their applications in chemosensors, biological imaging and white-light emitting materials are summarized. Most of the applications of the ESIPT chromophores are based on the photophysics properties, such as design of fluorescent chemosensors by perturbation of the ESIPT process upon interaction with the analytes, their use as biological fluorescent tags to study DNA–protein interaction by probing the variation of the hydration, or design of white-light emitting materials by employing the large Stokes shift of the ESIPT chromophores (to inhibit the Foster energy transfer of the components). The photophysical mechanism of these applications is discussed. Furthermore, a new research topic concerning the ESIPT chromophores is proposed based on our group's results, that is, to develop organic triplet sensitizers with ESIPT chromophores.

We report a ratiometric fluorescent sensor based on monochlorinated BODIPY for highly selective detection of glutathione (GSH) over cysteine (Cys)/homocysteine (Hcy). The chlorine of the monochlorinated BODIPY can be rapidly replaced by thiolates of biothiols through thiol–halogen nucleophilic substitution. The amino groups of Cys/Hcy but not GSH further replace the thiolate to form amino-substituted BODIPY. The significantly different photophysical properties of sulfur- and amino-substituted BODIPY enable the discrimination of GSH over Cys and Hcy. The sensor was applied for detection of GSH in living cells.

Design strategies of fluorescent probes for selective detection among biothiols

At elevated levels, homocysteine (Hcy, 1) is a risk factor for cardiovascular diseases, Alzheimer's disease, neural tube defects, and osteoporosis. Both 1 and cysteine (Cys, 3) are linked to neurotoxicity. The biochemical mechanisms by which 1 and 3 are involved in disease states are relatively unclear. Herein, we describe simple methods for detecting either Hcy or Cys in the visible spectral region with the highest selectivity reported to date without using biochemical techniques or preparative separations. Simple methods and readily available reagents allow for the detection of Cys and Hcy in the range of their physiologically relevant levels. New HPLC postcolumn detection methods for biological thiols are reported. The potential biomedical relevance of the chemical mechanisms involved in the detection of 1 is described.

Detection of Homocysteine and Cysteine

The synthesis of new benzo[a]- and [b]xanthene dye frameworks is described. A unique benzo[a]xanthene, seminaphtho[a]fluorone (SNAFR-1), is studied in a variety of media. The optimization of solution parameters and excitation wavelengths allows SNAFR-1 to display red, green, and blue emission bands of approximately equal intensities and also to produce white light. Ratiometric red (anion) and green (neutral) emissions are observed upon varying solution pH. A pH-independent violet-blue emission band is due to the addition of nucleophiles to the benzylic carbon of SNAFR-1.

An organic white light-emitting fluorophore.

A facile synthetic route utilizing readily available reagents affords a series of regioisomerically pure xanthene dye derivatives. Advantages include relatively mild conditions and good to excellent yields. Nonpolar, highly crystalline intermediates are isolable by standard chromatographic techniques. The intermediates are in the requisite xanthene oxidation state, thus avoiding the need for relatively inefficient oxidation chemistry and/or harsh conditions. During the course of this work, a new boron-mediated 1,2-aryl migration reaction was discovered.
