Clinical validation of a Cas13-based assay for the detection of SARS-CoV-2 RNA.
Maturada Patchsung,Krittapas Jantarug,Archiraya Pattama,Kanokpol Aphicho,Surased Suraritdechachai,Piyachat Meesawat,Khomkrit Sappakhaw,Nattawat Leelahakorn,Theerawat Ruenkam,Thanakrit Wongsatit,Niracha Athipanyasilp,Bhumrapee Eiamthong,Benya Lakkanasirorat,Thitima Phoodokmai,Nootaree Niljianskul,Danaya Pakotiprapha,Sittinan Chanarat,Aimorn Homchan,Ruchanok Tinikul,Philaiwarong Kamutira,Kochakorn Phiwkaow,Sahachat Soithongcharoen,Chadaporn Kantiwiriyawanitch,Vinutsada Pongsupasa,Duangthip Trisrivirat,Juthamas Jaroensuk,Thanyaporn Wongnate,Somchart Maenpuen,Pimchai Chaiyen,Sirichai Kamnerdnakta,Jirawat Swangsri,Suebwong Chuthapisith,Yongyut Sirivatanauksorn,Chutikarn Chaimayo,Ruengpung Sutthent,Wannee Kantakamalakul,Julia Joung,Alim Ladha,Xin Jin,Jonathan S. Gootenberg,Omar O. Abudayyeh,Feng Zhang,Navin Horthongkham,Chayasith Uttamapinant +43 more
TL;DR: The specific high-sensitivity enzymatic reporter unlocking (SHERLOCK) assay detected severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) RNA with high sensitivity and specificity in hundreds of nasopharyngeal and throat swab samples collected at Siriraj Hospital in Thailand.
Journal ArticleDOI
Crystal structure of Bacillus stearothermophilus UvrA provides insight into ATP-modulated dimerization, UvrB interaction, and DNA binding.
Danaya Pakotiprapha,Yoshihiko Inuzuka,Brian R. Bowman,Geri F. Moolenaar,Nora Goosen,David Jeruzalmi,Gregory L. Verdine +6 more
TL;DR: This work observes that the nucleotide-binding sites are formed in an intramolecular fashion and are not at the dimer interface as is typically found in other ABC ATPases, and shows that one of these is required for interaction with UvrB, its partner in lesion recognition.
Journal ArticleDOI
Structure and mechanism of the UvrA–UvrB DNA damage sensor
TL;DR: The structure of the Geobacillus stearothermophilus UvrA–UvrB complex, the damage-sensor in bacterial NER and a new structure of UVRA are determined, showing that the conserved signature domain II ofUvrA mediates a nexus of contacts among Uvr a, UvrB and DNA.
Journal ArticleDOI
Application of WST-8 based colorimetric NAD(P)H detection for quantitative dehydrogenase assays
Kamonwan Chamchoy,Danaya Pakotiprapha,Pornpan Pumirat,Ubolsree Leartsakulpanich,Usa Boonyuen +4 more
TL;DR: The results suggest that the WST-8 assay is a sensitive and rapid method for determining NAD(P)H concentration and dehydrogenase enzyme activity, which can be further applied for the high-throughput screening of dehydrogenases.
Journal ArticleDOI
A Structural Model for the Damage-sensing Complex in Bacterial Nucleotide Excision Repair
TL;DR: The structure of the complex between the interaction domains of UvrA and UvrB is reported, suggesting a model for the complete damage-sensing complex of bacterial nucleotide excision repair.
