Journal ArticleDOI

Crystal structure of Bacillus stearothermophilus UvrA provides insight into ATP-modulated dimerization, UvrB interaction, and DNA binding.

TL;DR: This work observes that the nucleotide-binding sites are formed in an intramolecular fashion and are not at the dimer interface as is typically found in other ABC ATPases, and shows that one of these is required for interaction with UvrB, its partner in lesion recognition.
Journal ArticleDOI

Structure and mechanism of the UvrA–UvrB DNA damage sensor

TL;DR: The structure of the Geobacillus stearothermophilus UvrA–UvrB complex, the damage-sensor in bacterial NER and a new structure of UVRA are determined, showing that the conserved signature domain II ofUvrA mediates a nexus of contacts among Uvr a, UvrB and DNA.
Journal ArticleDOI

Application of WST-8 based colorimetric NAD(P)H detection for quantitative dehydrogenase assays

TL;DR: The results suggest that the WST-8 assay is a sensitive and rapid method for determining NAD(P)H concentration and dehydrogenase enzyme activity, which can be further applied for the high-throughput screening of dehydrogenases.
Journal ArticleDOI

A Structural Model for the Damage-sensing Complex in Bacterial Nucleotide Excision Repair

TL;DR: The structure of the complex between the interaction domains of UvrA and UvrB is reported, suggesting a model for the complete damage-sensing complex of bacterial nucleotide excision repair.
