Toward rational thermostabilization of Aspergillus oryzae cutinase: Insights into catalytic and structural stability.

TL;DR: Three notable findings are presented as steps toward designing more thermophilic cutinase: surface salt bridge optimization produced enthalpic stabilization, mutations to proline reduced the entropy loss upon folding, and the lack of a correlative increase in the temperature optimum of catalytic activity with thermodynamic stability suggests that the active site is locally denatured at a temperature below the Tm of the global structure.

Influence of surface charge, binding site residues and glycosylation on Thielavia terrestris cutinase biochemical characteristics

TL;DR: The stability of TtC in acidic pH was rationalized by evaluating the contribution of charge interactions to the Gibbs free energy of unfolding at varying pHs and the activity of TTC was found to be limited by substrate binding affinity, which is a function of the surface charge.
