TL;DR: In this paper, the authors show that lysosomes, sophisticated nutrient-sensing and signaling centers, are regulated dichotomously by transcription factor EB (TFEB) and MYC to balance catabolic and anabolic processes required for activating LT-HSCs and guiding their lineage fate.
Journal ArticleDOI
3232- – inflammatory stress promotes heterogeneous activation of immune response programs in human hematopoietic stem cells
Stephanie Z. Xie,Murtaza S. Nagree,Andy G.X. Zeng,Alex Murison,Joana Araujo,Isabel Lim,Darrien Parris,Jessica McLeod,Liqing Jin,Elvin Wagenblast,John E. Dick +10 more
TL;DR: In this article , a TNFα-mediated inflammatory challenge xenotransplantation model was used to study the effect of TNF on the progression from quiescence to cellular activation in LT-HSC.
Journal ArticleDOI
Discovery of a New Human Hematopoietic Stem Cell Involved with Aging That Retains Memory of Immune Activation
Andy G.X. Zeng,Murtaza S. Nagree,Alex Murison,Isabel Lim,Sayyam Shah,Alicia G. Aguilar-Navarro,Joana Araujo,Darrien Parris,Jessica McLeod,Liqing Jin,Elvin Wagenblast,Eugenia Flores-Figueroa,John E. Dick,Stephanie Z. Xie +13 more
TL;DR: In this paper , the authors observed a gradient of TNFα-via-NFkB pathway enrichment in single cell (sc) transcriptomes of ~4000 cord blood (CB) LT-HSC.
