Journal ArticleDOI
Chromatin extrusion explains key features of loop and domain formation in wild-type and engineered genomes.
Adrian L. Sanborn,Adrian L. Sanborn,Adrian L. Sanborn,Suhas S.P. Rao,Suhas S.P. Rao,Su Chen Huang,Neva C. Durand,Miriam H. Huntley,Andrew I. Jewett,Ivan D. Bochkov,Dharmaraj Chinnappan,Ashok Cutkosky,Jian Li,Jian Li,Kristopher Geeting,Andreas Gnirke,Alexandre Melnikov,Doug McKenna,Elena K. Stamenova,Elena K. Stamenova,Eric S. Lander,Eric S. TL;DR: It is shown that it is possible to disrupt, restore, and move loops and domains using targeted mutations as small as a single base pair at CTCF sites, and it is found that the observed contact domains are inconsistent with the equilibrium state for an ordinary condensed polymer.
Journal ArticleDOI
Abundance of two human preadipocyte subtypes with distinct capacities for replication, adipogenesis, and apoptosis varies among fat depots.
Tamara Tchkonia,Yourka D. Tchoukalova,Nino Giorgadze,Tamar Pirtskhalava,Iordanes Karagiannides,R. Armour Forse,Ada Koo,Michael Stevenson,Dharmaraj Chinnappan,Andrew Cartwright,Michael D. Jensen,James L. Kirkland +11 more
TL;DR: In this article, the preadipocytes that fat cells develop from exhibit distinct regional characteristics that persist in culture, and human abdominal subcutaneous cultured pre-adipo...
Journal ArticleDOI
Fat depot-specific characteristics are retained in strains derived from single human preadipocytes.
Tamara Tchkonia,Nino Giorgadze,Tamar Pirtskhalava,Thomas Thomou,Matthew DePonte,Ada Koo,R. Armour Forse,Dharmaraj Chinnappan,Carmen Martin-Ruiz,Thomas von Zglinicki,James L. Kirkland +10 more
TL;DR: Despite dividing 40 population doublings, hTERT strains derived from single preadipocytes retained fat depot–specific cell dynamic characteristics, consistent with heritable processes contributing to regional variation in fat tissue function.
Journal ArticleDOI
Mechanism of Aurora-B degradation and its dependency on intact KEN and A-boxes: identification of an aneuploidy-promoting property.
TL;DR: Evidence is provided that Aurora-B is a short-lived protein degraded by the proteasome via the anaphase-promoting cyclosome complex (APC/c) pathway, and overexpression of a stable form of Aurora- B alone induces aneuploidy and anchorage-independent growth.
Journal ArticleDOI
Differential DNA Hypermethylation of Critical Genes Mediates the Stage-Specific Tobacco Smoke-Induced Neoplastic Progression of Lung Cancer
Andrea L. Russo,Arunthathi Thiagalingam,Hongjie Pan,Joseph A. Califano,Kuang-hung Cheng,Jose F. Ponte,Dharmaraj Chinnappan,Pratima Nemani,David Sidransky,Sam Thiagalingam +9 more
TL;DR: The data suggest that targeted DNA methylation silencing of ECAD and DAPK occurs in the early stages and that of p16 and MGMT in the later stages of lung cancer progression, and provides preliminary evidence that peripheral lymphocytes could potentially be used as a surrogate for bronchial epithelial cells to detect alteredDNA methylation in smokers.
