Journal ArticleDOI
Mitochondrial impairment in microglia amplifies NLRP3 inflammasome proinflammatory signaling in cell culture and animal models of Parkinson's disease.
Souvarish Sarkar,Emir Malovic,Dilshan S. Harishchandra,Dilshan S. Harishchandra,Shivani Ghaisas,Shivani Ghaisas,Nikhil Panicker,Nikhil Panicker,Adhithiya Charli,Bharathi N. Palanisamy,Dharmin Rokad,Huajun Jin,Vellareddy Anantharam,Arthi Kanthasamy,Anumantha G. Kanthasamy +14 more
TL;DR: It is shown that impaired mitochondrial function can augment the NLRP3 inflammasome-driven proinflammatory cascade in microglia and contribute to better understand the mechanisms driving chronic neuroinflammation in PD.
Journal ArticleDOI
Fyn kinase regulates misfolded α-synuclein uptake and NLRP3 inflammasome activation in microglia.
Nikhil Panicker,Nikhil Panicker,Souvarish Sarkar,Dilshan S. Harischandra,Matthew Neal,Tae In Kam,Huajun Jin,Hariharan Saminathan,Monica R. Langley,Adhithiya Charli,Manikandan Samidurai,Dharmin Rokad,Shivani Ghaisas,Olga Pletnikova,Valina L. Dawson,Ted M. Dawson,Vellareddy Anantharam,Anumantha G. Kanthasamy,Arthi Kanthasamy +18 more
TL;DR: Fyn kinase mediates aggregated α-synuclein uptake as well as αSyn-mediated proinflammatory signaling in microglia, leading to NLRP3 inflammasome activation and neuroinflammation in Parkinson’s disease.
Journal ArticleDOI
Manganese promotes the aggregation and prion-like cell-to-cell exosomal transmission of α-synuclein
Dilshan S. Harischandra,Dharmin Rokad,Matthew Neal,Shivani Ghaisas,Sireesha Manne,Souvarish Sarkar,Nikhil Panicker,Gary Zenitsky,Huajun Jin,Mechelle M. Lewis,Xuemei Huang,Vellareddy Anantharam,Arthi Kanthasamy,Anumantha G. Kanthasamy +13 more
TL;DR: Results indicate that Mn2+ exposure promotes αSyn secretion in exosomal vesicles, which subsequently evokes proinflammatory and neurodegenerative responses in both cell culture and animal models.
Journal ArticleDOI
Manganese exposure induces neuroinflammation by impairing mitochondrial dynamics in astrocytes
Souvarish Sarkar,Emir Malovic,Dilshan S. Harischandra,Hilary Afeseh Ngwa,Anamitra Ghosh,Colleen Hogan,Dharmin Rokad,Gary Zenitsky,Huajun Jin,Vellareddy Anantharam,Anumantha G. Kanthasamy,Arthi Kanthasamy +11 more
TL;DR: It is demonstrated that Mn drives proinflammatory events in astrocytes by impairing mitochondrial bioenergetics and the novel mitochondria‐targeted antioxidant, mito‐apocynin, significantly attenuated Mn‐induced inflammatory gene expression, further supporting the role of mitochondria dysfunction and oxidative stress in mediating astrogliosis.
Journal ArticleDOI
Manganese activates NLRP3 inflammasome signaling and propagates exosomal release of ASC in microglial cells
Souvarish Sarkar,Dharmin Rokad,Emir Malovic,Jie Luo,Dilshan S. Harischandra,Huajun Jin,Vellareddy Anantharam,Xuemei Huang,Mechelle M. Lewis,Arthi Kanthasamy,Anumantha G. Kanthasamy +10 more
TL;DR: Results suggest that the divalent metal manganese acts as a key amplifier of NLRP3 inflammasome signaling and exosomal ASC release.
