Chlamydophila psittaci infections in birds: a review with emphasis on zoonotic consequences.

Evidence for the existence of two new members of the family Chlamydiaceae and proposal of Chlamydia avium sp. nov. and Chlamydia gallinacea sp. nov.

Wild waterfowl, particularly dabbling ducks such as mallards (Anas platyrhynchos), are considered the main reservoir of low-pathogenic avian influenza viruses (LPAIVs). They carry viruses that may evolve and become highly pathogenic for poultry or zoonotic. Understanding the ecology of LPAIVs in these natural hosts is therefore essential. We assessed the clinical response, viral shedding and antibody production of juvenile mallards after intra-esophageal inoculation of two LPAIV subtypes previously isolated from wild congeners. Six ducks, equipped with data loggers that continually monitored body temperature, heart rate and activity, were successively inoculated with an H7N7 LPAI isolate (day 0), the same H7N7 isolate again (day 21) and an H5N2 LPAI isolate (day 35). After the first H7N7 inoculation, the ducks remained alert with no modification of heart rate or activity. However, body temperature transiently increased in four individuals, suggesting that LPAIV strains may have minor clinical effects on their natural hosts. The excretion patterns observed after both re-inoculations differed strongly from those observed after the primary H7N7 inoculation, suggesting that not only homosubtypic but also heterosubtypic immunity exist. Our study suggests that LPAI infection has minor clinically measurable effects on mallards and that mallard ducks are able to mount immunological responses protective against heterologous infections. Because the transmission dynamics of LPAIVs in wild populations is greatly influenced by individual susceptibility and herd immunity, these findings are of high importance. Our study also shows the relevance of using telemetry to monitor disease in animals.

Influenza Virus in a Natural Host, the Mallard: Experimental Infection Data

Psittacosis, also known as parrot fever and ornithosis, is a bacterial infection that can cause severe pneumonia and other serious health problems in humans. It is caused by Chlamydia psittaci. Reclassification of the order Chlamydiales in 1999 into 2 genera (Chlamydia and Chlamydophila) was not wholly accepted or adopted. This resulted in a reversion to the single, original genus Chlamydia, which now encompasses all 9 species including Chlamydia psittaci. During 2003–2014, 112 human cases of psittacosis were reported to the Centers for Disease Control and Prevention through the Nationally Notifiable Diseases Surveillance System. While many types of birds can be infected by C psittaci, in general, the literature suggests that human cases can most often occur after exposure to infected parrot-type birds kept as pets, especially cockatiels, parakeets, and conures. In birds, C psittaci infection is referred to as avian chlamydiosis. Infected birds shed the bacteria through feces and nasal discharges, and humans become infected from exposure to these materials. This compendium provides information about psittacosis and avian chlamydiosis to public health officials, physicians, veterinarians, the pet bird industry, and others concerned with controlling these diseases and protecting public health. The recommendations in this compendium provide standardized procedures to control C psittaci infections. This document will be reviewed and revised as necessary, and the most current version replaces all previous versions. This document was last revised in 2010. Major changes in this version include a recommendation for a shorter treatment time for birds with avian chlamydiosis, additional information about diagnostic testing, including genotyping, clearer language associated with personal protective equipment recommended for those caring for confirmed or exposed birds, and incorporating a grading scale with recommendations generally based on the United States Preventive Services Task Force's methods.

Compendium of Measures to Control Chlamydia psittaci Infection Among Humans (Psittacosis) and Pet Birds (Avian Chlamydiosis), 2017.

Chlamydia comprises a group of obligate intracellular bacterial parasites responsible for a variety of diseases in humans and animals, including several zoonoses. Chlamydia trachomatis causes diseases such as trachoma, urogenital infection and lymphogranuloma venereum with severe morbidity. Chlamydia pneumoniae is a common cause of community-acquired respiratory tract infections. Chlamydia psittaci, causing zoonotic pneumonia in humans, is usually hosted by birds, while Chlamydia abortus, causing abortion and fetal death in mammals, including humans, is mainly hosted by goats and sheep. We used multi-locus sequence typing to asses the population structure of Chlamydia. In total, 132 Chlamydia isolates were analyzed, including 60 C. trachomatis, 18 C. pneumoniae, 16 C. abortus, 34 C. psittaci and one of each of C. pecorum, C. caviae, C. muridarum and C. felis. Cluster analyses utilizing the Neighbour-Joining algorithm with the maximum composite likelihood model of concatenated sequences of 7 housekeeping fragments showed that C. psittaci 84/2334 isolated from a parrot grouped together with the C. abortus isolates from goats and sheep. Cluster analyses of the individual alleles showed that in all instances C. psittaci 84/2334 formed one group with C. abortus. Moving 84/2334 from the C. psittaci group to the C. abortus group resulted in a significant increase in the number of fixed differences and elimination of the number of shared mutations between C. psittaci and C. abortus. C. psittaci M56 from a muskrat branched separately from the main group of C. psittaci isolates. C. psittaci genotypes appeared to be associated with host species. The phylogentic tree of C. psittaci did not follow that of its host bird species, suggesting host species jumps. In conclusion, we report for the first time an association between C. psittaci genotypes with host species.

The diagnosis of Chlamydia trachomatis infection can be based either on direct detection of the organism or its components or indirectly by measuring antibodies as markers of the individual's response to the infection. The latter is currently of limited value. Neither IgG or IgA antibodies can be used to diagnose current genital infection by Chlamydia trachomatis or to exclude such an infection. There is no solid ground as yet for the use of IgA antibodies as a marker of persistant or unresolved infection. Commercial tests in the Elisa format based on peptides from the MOMP of Chlamydia trachomatis are available and show good specificities and sensitivities. Hsp60 seems to have a unique role in the development of tubal scarring and antibodies to chsp60 could predict tubal factor infertility. Serology is the main diagnostic tool for the diagnosis of Mycoplasma pneumoniae infection. The serologic assays are the complement fixation test (CF), immunofluorescence, the microparticle agglutination and recently EIAs. The CF test is still used for serodiagnosis of Mycoplasma pneumoniae infection because of the sensitivity of 90%. Single titer of >or= 64 are considered to be indicative of recent infection. A number of commercial EIAs have been developped. The difficulty for IgG interpretation is a definition of a cutoff value for discriminating infected and healthy subjects. Most of the IgM assays show good diagnostic sensitivities and are valuable tools for the early diagnosis of Mycoplasma pneumoniae infection in children. There are no wholly satisfactory serological methods for diagnosis of Chlamydia pneumoniae infection. Problems arise from the high background of IgG antibody prevalence, the lack of standardized testing methods.

Resume Trois especes sont responsables de chlamydioses chez l’homme : Chlamydia trachomatis , principal agent bacterien d’infection sexuellement transmissible, Chlamydia pneumoniae , responsable d’infection respiratoire communautaire et Chlamydia psittaci , d’origine aviaire, occasionnellement responsable de pneumopathie. Le diagnostic d’infection a C. trachomatis repose essentiellement sur la recherche directe par amplification genique avec des outils commercialises, sensibles et specifiques. Le serodiagnostic a un interet dans l’evaluation de l’extension de la maladie. Le diagnostic d’infection a C. pneumoniae a beaucoup souffert du manque de specificite des techniques utilisees apres sa decouverte. Il semble que l’incidence de l’infection respiratoire a C. pneumoniae ait ete largement surestimee. Dans l’etat actuel de nos connaissances, les techniques serologiques pourraient etre abandonnees au profit de techniques directes telles que l’amplification genique avec des trousses commercialisees, sensibles et specifiques, en duplex avec Mycoplasma pneumoniae . Quant a C. psittaci , son diagnostic releve de laboratoire specialise, en raison de l’absence de techniques directes commercialisees disponibles et de l’expertise requise pour interpreter le serodiagnostic.
