Stoichiometry and turnover in single, functioning membrane protein complexes
Mark C. Leake,Jennifer H. Chandler,George H. Wadhams,Fan Bai,Richard M. Berry,Judith P. Armitage +5 more
TL;DR: The number and rapid turnover of protein subunits within a functioning molecular machine are measured with single-molecule precision in functioning bacterial flagellar motors in Escherichia coli for the first time.
Journal ArticleDOI
Genomic and Transcriptomic Landscape of Triple-Negative Breast Cancers: Subtypes and Treatment Strategies.
Yi-Zhou Jiang,Ding Ma,Chen Suo,Shi Jinxiu,Mengzhu Xue,Xin Hu,Yi Xiao,Ke Da Yu,Yi Rong Liu,Ying Yu,Yuanting Zheng,Xiangnan Li,Chen-Hui Zhang,Peng-Chen Hu,Jing Zhang,Qi Hua,Jiyang Zhang,Wanwan Hou,Luyao Ren,Ding Bao,B. Kaklamani,Anneleen Daemen,John R. Benson,Kazuaki Takabe,Fan Bai,Da Qiang Li,Peng Wang,Leming Shi,Wei Huang,Zhi Ming Shao +42 more
TL;DR: The LAR subtype showed more ERBB2 somatic mutations, infrequent mutational signature 3 and frequent CDKN2A loss than the other subtypes, and was classified as a mesenchymal-like subtype.
Journal ArticleDOI
Single-cell landscape of immunological responses in patients with COVID-19.
Ji Yuan Zhang,Xiang Ming Wang,Xudong Xing,Zhe Xu,Chao Zhang,Jin-Wen Song,Xing Fan,Peng Xia,Jun Liang Fu,Si Yu Wang,Ruo Nan Xu,Xiao Peng Dai,Lei Shi,Lei Huang,Tian Jun Jiang,Ming Shi,Yuxia Zhang,Alimuddin Zumla,Alimuddin Zumla,Markus Maeurer,Fan Bai,Fu-Sheng Wang +21 more
TL;DR: The single-cell transcriptional landscape of moderate, severe and convalescent cases of patients with COVID-19 describes the dynamic nature of immune responses during disease progression, and shows a deranged interferon response, profound immune exhaustion with skewed T cell receptor repertoire and broad T cell expansion.
Journal ArticleDOI
TGF-β–induced epithelial-to-mesenchymal transition proceeds through stepwise activation of multiple feedback loops
Jingyu Zhang,Jingyu Zhang,Xiao-Jun Tian,Hang Zhang,Yue Teng,Ruoyan Li,Fan Bai,Subbiah Elankumaran,Jianhua Xing +8 more
TL;DR: Experimental and computational analyses identify multiple cell populations during the epithelial-to-mesenchymal transition and provide experimental confirmation for a model of cascading switches in phenotypes associated with TGF-β1–induced EMT of MCF10A cells that involves two double-negative feedback loops.
Journal ArticleDOI
Reproducible copy number variation patterns among single circulating tumor cells of lung cancer patients
Xiaohui Ni,Xiaohui Ni,Minglei Zhuo,Zhe Su,Jianchun Duan,Yan Gao,Zhijie Wang,Chenghang Zong,Hua Bai,Alec R. Chapman,Jun Zhao,Liya Xu,Tongtong An,Qi Ma,Yuyan Wang,Meina Wu,Yu Sun,Shuhang Wang,Zhenxiang Li,Xiaodan Yang,Jun Yong,Xiao-Dong Su,Youyong Lu,Fan Bai,X. Sunney Xie,X. Sunney Xie,Jie Wang +26 more
TL;DR: The use of the recently developed multiple annealing and looping-based amplification cycles for whole-genome amplification of single CTCs from lung cancer patients are reported, finding that copy number variations (CNVs), one of the major genomic variations, are specific to cancer types, reproducible from cell to cell, and even from patient to patient.
