Journal ArticleDOI
Slow- and tight-binding inhibitors of the 85-kDa human phospholipase A2.
Ian P. Street,Hung-Kuei Lin,F. Laliberte,Farideh Ghomashchi,Zhaoyin Wang,Helene Perrier,Nathalie Tremblay,Zheng Huang,Philip K. Weech,Michael H. Gelb +9 more
TL;DR: A trifluoromethyl ketone analogue of arachidonic acid in which the COOH group is replaced with COCF3 was prepared and found to be a tight- and slow-binding inhibitor of the 85-kDa cytosolic human phospholipase A2 (cPLA2).
Journal ArticleDOI
Interfacial Kinetic and Binding Properties of the Complete Set of Human and Mouse Groups I, II, V, X, and XII Secreted Phospholipases A2
Alan G. Singer,Farideh Ghomashchi,Catherine Le Calvez,James G. Bollinger,Sofiane Bezzine,Morgane Rouault,Martin Sadilek,Eric Nguyen,Michel Lazdunski,Gérard Lambeau,Michael H. Gelb +10 more
TL;DR: A dramatic correlation exists between the ability of the sPLA2s to hydrolyze phosphatidylcholine-rich vesicles efficiently in vitro and the ability to release arachidonic acid when added exogenously to mammalian cells; the group V and X s PLA2s are uniquely efficient in this regard.
Journal ArticleDOI
Serine 727 phosphorylation and activation of cytosolic phospholipase A2 by MNK1-related protein kinases.
Ying Hefner,Angelika G. Börsch-Haubold,Makomoto Murakami,Jonathan I. Wilde,Sophie Pasquet,David Schieltz,Farideh Ghomashchi,John R. Yates,Christopher G. Armstrong,Andrew D. Paterson,Philip Cohen,Rikiro Fukunaga,Tony Hunter,Ichiro Kudo,Steve P. Watson,Michael H. Gelb +15 more
TL;DR: It is shown that phosphorylation of cPLA2 at both Ser-505 and Ser-727 and elevation of Ca2+ leads to its activation in agonist-stimulated cells and suggests that MNK1 or a closely related kinase is responsible for in vivo phosphorylated of c PLA2 onSer-727.
Journal ArticleDOI
A protein geranylgeranyltransferase from bovine brain: implications for protein prenylation specificity.
TL;DR: Results suggest that the C-terminal amino acid is an important structural determinant in controlling the specificity of protein prenylation.
Journal ArticleDOI
Exogenously Added Human Group X Secreted Phospholipase A2 but Not the Group IB, IIA, and V Enzymes Efficiently Release Arachidonic Acid from Adherent Mammalian Cells
Sofiane Bezzine,Rao S. Koduri,Emmanuel Valentin,Makoto Murakami,Ichiro Kudo,Farideh Ghomashchi,Martin Sadilek,Gérard Lambeau,Michael H. Gelb +8 more
TL;DR: Studies with CHO-K1 cell mutants show that binding of sPLA2s to glycosaminoglycans is not the basis for poor plasma membrane hydrolysis by group IB, IIA, and V sPLA 2s.
