RNA methylation by Dnmt2 protects transfer RNAs against stress-induced cleavage
Matthias Schaefer,Tim Pollex,Katharina Hanna,Francesca Tuorto,Madeleine Meusburger,Madeleine Meusburger,Mark Helm,Mark Helm,Frank Lyko +8 more
TL;DR: Results uncover a novel biological function of Dnmt2-mediated tRNA methylation, and suggest a role for DnMT2 enzymes during the biogenesis of tRNA-derived small RNAs.
Journal ArticleDOI
RNA cytosine methylation by Dnmt2 and NSun2 promotes tRNA stability and protein synthesis.
Francesca Tuorto,Reinhard Liebers,Tanja Musch,Matthias Schaefer,Sarah Hofmann,Stefanie Kellner,Michaela Frye,Mark Helm,Georg Stoecklin,Frank Lyko +9 more
TL;DR: A biologically important function for cytosine-C5 tRNA methylation in mammals is established and it is suggested that this modification promotes mouse development by supporting protein synthesis.
Journal ArticleDOI
RNA cytosine methylation analysis by bisulfite sequencing
TL;DR: A protocol for the chemical deamination of cytosines in RNA is established, followed by PCR-based amplification of cDNA and DNA sequencing, and it is shown that cytosine methylation can be reproducibly and quantitatively detected by bisulfite sequencing.
Journal ArticleDOI
Loss of Tet Enzymes Compromises Proper Differentiation of Embryonic Stem Cells
Meelad M. Dawlaty,Achim Breiling,Thuc Le,M. Inmaculada Barrasa,Günter Raddatz,Qing Gao,Benjamin E. Powell,Albert W. Cheng,Kym F. Faull,Frank Lyko,Rudolf Jaenisch +10 more
TL;DR: In this paper, the effect of Tet1/2/3 triple-knockout (TKO) mouse embryonic stem cells (ESCs) on ESC development was examined.
Loss of Tet Enzymes Compromises Proper Differentiation of Embryonic Stem Cells
Meelad M. Dawlaty,Achim Breiling,Thuc Le,M. Inmaculada Barrasa,Günter Raddatz,Qing Gao,Benjamin E. Powell,Albert W. Cheng,Kym F. Faull,Frank Lyko,Rudolf Jaenisch +10 more
TL;DR: A requirement for Tet- and 5hmC-mediated DNA demethylation in proper regulation of gene expression during ESC differentiation and development is suggested.
