A paradigm shift is taking place in medicine from using synthetic implants and tissue grafts to a tissue engineering approach that uses degradable porous material scaffolds integrated with biological cells or molecules to regenerate tissues. This new paradigm requires scaffolds that balance temporary mechanical function with mass transport to aid biological delivery and tissue regeneration. Little is known quantitatively about this balance as early scaffolds were not fabricated with precise porous architecture. Recent advances in both computational topology design (CTD) and solid free-form fabrication (SFF) have made it possible to create scaffolds with controlled architecture. This paper reviews the integration of CTD with SFF to build designer tissue-engineering scaffolds. It also details the mechanical properties and tissue regeneration achieved using designer scaffolds. Finally, future directions are suggested for using designer scaffolds with in vivo experimentation to optimize tissue-engineering treatments, and coupling designer scaffolds with cell printing to create designer material/biofactor hybrids.

Porous scaffold design for tissue engineering

In this paper we develop a new constitutive law for the description of the (passive) mechanical response of arterial tissue. The artery is modeled as a thick-walled nonlinearly elastic circular cylindrical tube consisting of two layers corresponding to the media and adventitia (the solid mechanically relevant layers in healthy tissue). Each layer is treated as a fiber-reinforced material with the fibers corresponding to the collagenous component of the material and symmetrically disposed with respect to the cylinder axis. The resulting constitutive law is orthotropic in each layer. Fiber orientations obtained from a statistical analysis of histological sections from each arterial layer are used. A specific form of the law, which requires only three material parameters for each layer, is used to study the response of an artery under combined axial extension, inflation and torsion. The characteristic and very important residual stress in an artery in vitro is accounted for by assuming that the natural (unstressed and unstrained) configuration of the material corresponds to an open sector of a tube, which is then closed by an initial bending to form a load-free, but stressed, circular cylindrical configuration prior to application of the extension, inflation and torsion. The effect of residual stress on the stress distribution through the deformed arterial wall in the physiological state is examined. The model is fitted to available data on arteries and its predictions are assessed for the considered combined loadings. It is explained how the new model is designed to avoid certain mechanical, mathematical and computational deficiencies evident in currently available phenomenological models. A critical review of these models is provided by way of background to the development of the new model.

A new constitutive framework for arterial wall mechanics and a comparative study of material models

Constitutive relations are fundamental to the solution of problems in continuum mechanics, and are required in the study of, for example, mechanically dominated clinical interventions involving soft biological tissues. Structural continuum constitutive models of arterial layers integrate information about the tissue morphology and therefore allow investigation of the interrelation between structure and function in response to mechanical loading. Collagen fibres are key ingredients in the structure of arteries. In the media (the middle layer of the artery wall) they are arranged in two helically distributed families with a small pitch and very little dispersion in their orientation (i.e. they are aligned quite close to the circumferential direction). By contrast, in the adventitial and intimal layers, the orientation of the collagen fibres is dispersed, as shown by polarized light microscopy of stained arterial tissue. As a result, continuum models that do not account for the dispersion are not able to capture accurately the stress–strain response of these layers. The purpose of this paper, therefore, is to develop a structural continuum framework that is able to represent the dispersion of the collagen fibre orientation. This then allows the development of a new hyperelastic free-energy function that is particularly suited for representing the anisotropic elastic properties of adventitial and intimal layers of arterial walls, and is a generalization of the fibre-reinforced structural model introduced by Holzapfel & Gasser (Holzapfel & Gasser 2001 Comput. Meth. Appl. Mech. Eng. 190, 4379–4403) and Holzapfel et al. (Holzapfel et al. 2000 J. Elast. 61, 1–48). The model incorporates an additional scalar structure parameter that characterizes the dispersed collagen orientation. An efficient finite element implementation of the model is then presented and numerical examples show that the dispersion of the orientation of collagen fibres in the adventitia of human iliac arteries has a significant effect on their mechanical response.

The experimental quantification and modeling of the multiaxial mechanical response of polymer membranes of coronary balloon catheters have not yet been carried out Due to the lack of insights, it is not shown whether isotropic material models can describe the material response of balloon catheter membranes expanded with nominal or higher, supra-nominal pressures Therefore, for the first time, specimens of commercial polyamide-12 balloon catheters membranes were investigated during uniaxial and biaxial loading scenarios Furthermore, the influence of kinematic effects on the material response was observed by comparing results from quasi-static and dynamic biaxial extension tests Novel clamping techniques are described, which allow to test even tiny specimens taken from the balloon membranes The results of this study reveal the semi-compliant, nonlinear, and viscoelastic character of polyamide-12 balloon catheter membranes Above nominal pressure, the membranes show a pronounced anisotropic mechanical behavior with a stiffer response in the circumferential direction The anisotropic feature intensifies with an increasing strain-rate A modified polynomial model was applied to represent the realistic mechanical response of the balloon catheter membranes during dynamic biaxial extension tests This study also includes a compact set of constitutive model parameters for the use of the proposed model in future finite element analyses to perform more accurate simulations of expanding balloon catheters

Computer Models in Biomechanics: From Nano to Macro

Compliance mismatch between the graft and the host artery of an end-to-side (ETS) arterial bypass graft anastomosis increases the intramural stress in the ETS graft-artery junction, and thus may compromise its long-term patency. The present study takes into account the effects of collagen fibers to demonstrate how their orientations alter the stresses. The stresses in an ETS bypass graft anastomosis, as a man-made bifurcation, are compared to those of its natural counterpart with different fiber orientations. Both of the ETS bypass graft anastomosis and its natural counterpart have identical geometric and material models and only their collagen fiber orientations are different. The results indicate that the fiber orientation mismatch between the graft and the host artery may increase the stresses at both the heel and toe regions of the ETS anastomosis (the maximum principal stress at the heel and toe regions increased by 72% and 12%, respectively). Our observations, thus, propose that the mismatch between the collagen fiber orientations of the graft and the host artery, independent of the effect of the suture line, may induce aberrant stresses to the anastomosis of the bypass graft.

Role of Vessel Microstructure in the Longevity of End-to-side Grafts.

Classical continuum mechanics has been widely used for implementation of the material models of articular cartilage (AC) mainly with the aid of the finite element (FE) method, which, in many cases, considers the stress-free configuration as the initial configuration. On the contrary, the AC experimental tests typically begin with the pre-stressed state of both material and geometrical properties. Indeed, imposing the initial pre-stress onto AC models with the in vivo values as the initial state would result in nonphysiologically expansion of the FE mesh due to the soft nature of AC. This change in the model configuration can also affect the material behavior kinematically in the mixture models of cartilage due to the intrinsic compressibility of the tissue. Although several different fixed-point backward algorithms, as the most straightforward pre-stressing methods, have already been developed to incorporate these initial conditions into FE models iteratively, such methods focused merely on the geometrical parameters, and they omitted the material variations of the anisotropic mixture models of AC. To address this issue, we propose an efficient algorithm generalizing the backward schemes to restore stress-free conditions by optimizing both the involving variables, and we hypothesize that it can affect the results considerably. To this end, a comparative simulation was implemented on an advanced and validated multiphasic model by the new and conventional algorithms. The results are in support of the hypothesis, as in our illustrative general AC model, the material parameters experienced a maximum error of 16% comparing to the initial in vivo data when the older algorithm was employed, and it led to a maximum variation of 44% in the recorded stresses comparing to the results of the new method. We conclude that our methodology enhanced the model fidelity, and it is applicable in most of the existing FE solvers for future mixture studies with accurate stress distributions.
