Autophagy deficiency leads to protection from obesity and insulin resistance by inducing Fgf21 as a mitokine
Kook Hwan Kim,Yeon Taek Jeong,Hyunhee Oh,Seonghun Kim,Jae Min Cho,Yo-Na Kim,Su Sung Kim,Dohoon Kim,Kyu Yeon Hur,Hyoung Kyu Kim,Tae-Hee Ko,Jin Han,Hong Lim Kim,Jin Kim,Sung Hoon Back,Masaaki Komatsu,Hsiuchen Chen,David C. Chan,Morichika Konishi,Nobuyuki Itoh,Cheol Soo Choi,Myung-Shik Lee,Myung-Shik Lee +22 more
TL;DR: It is suggested that autophagy deficiency and subsequent mitochondrial dysfunction promote Fgf21 expression, a hormone the authors consequently term a 'mitokine', and together these processes promote protection from diet-induced obesity and insulin resistance.
Journal ArticleDOI
Systemic autophagy insufficiency compromises adaptation to metabolic stress and facilitates progression from obesity to diabetes.
Yu Mi Lim,H.Y. Lim,Kyu Yeon Hur,Wenying Quan,Hae Youn Lee,Hwanju Cheon,Dongryeol Ryu,Seung Hoi Koo,Hong Lim Kim,Jin Kim,Masaaki Komatsu,Myung-Shik Lee +11 more
TL;DR: It is shown that mice with global haploinsufficiency of an essential autophagy gene (Atg7(+/-) mice) do not show metabolic abnormalities but develop diabetes when crossed with ob/ob mice, suggesting that systemic autophagic insufficiency could be a factor in the progression from obesity to diabetes.
Journal ArticleDOI
The role of autophagy in unilateral ureteral obstruction rat model.
Wan-Young Kim,Sun Ah Nam,Ho Cheol Song,Jun Sung Ko,Sang Hee Park,Hong Lim Kim,Euy Jin Choi,Yong Soo Kim,Jin Kim,Yong Kyun Kim +9 more
TL;DR: This work investigates the role of autophagy in the obstructed kidney and contralateral kidney after UUO, a model of progressive renal fibrosis in the Obstructed kidney that is followed by compensatory cellular proliferation in the contralsateral kidney.
Journal ArticleDOI
Autophagy attenuates tubulointerstital fibrosis through regulating transforming growth factor-β and NLRP3 inflammasome signaling pathway.
Sun Ah Nam,Wan Young Kim,Jin Won Kim,Sang Hee Park,Hong Lim Kim,Myung-Shik Lee,Masaaki Komatsu,Hunjoo Ha,Ji Hee Lim,Cheol Whee Park,Chul Woo Yang,Jin Kim,Yong Kyun Kim +12 more
TL;DR: The data showed that autophagy in distal TEC plays a protective role in development of renal tubulointerstial fibrosis through regulating the expression of TGF-β an IL1-β after UUO.
Journal ArticleDOI
Graft immaturity and safety concerns in transplanted human kidney organoids
Sun Ah Nam,Eunjeong Seo,Jin Won Kim,Hyung Wook Kim,Hong Lim Kim,Kyuryung Kim,Tae Min Kim,Ji Hyeon Ju,Ivan G. Gomez,Kohei Uchimura,Benjamin D. Humphreys,Chul Woo Yang,Jae Yeon Lee,Jin Kim,Dong-Woo Cho,Benjamin S. Freedman,Yong Kyun Kim +16 more
TL;DR: The data suggest that kidney organoids derived from iPS cells may be transplantable but strategies to improve nephron differentiation and purity are required before they can be applied in humans as a therapeutic option.
