Journal ArticleDOI
Crystal structure of Cas9 in complex with guide RNA and target DNA
Takashi Yamano,Hiroshi Nishimasu,Hiroshi Nishimasu,Bernd Zetsche,Hisato Hirano,Ian Slaymaker,Yinqing Li,Iana Fedorova,Takanori Nakane,Kira S. Makarova,Eugene V. Koonin,Ryuichiro Ishitani,Feng Zhang,Osamu Nureki +13 more
TL;DR: In this article, the crystal structure of the CRISPR-associated endonuclease Cas9 in complex with sgRNA and its target DNA at 2.5-A resolution was reported.
Journal ArticleDOI
Type III CRISPR-Cas Systems: Deciphering the Most Complex Prokaryotic Immune System
Matvey Kolesnik,Iana Fedorova,Iana Fedorova,Karyna A. Karneyeva,Daria Artamonova,Konstantin Severinov,Konstantin Severinov +6 more
TL;DR: A comprehensive review of Type III CRISPR-Cas components, their biological roles, and evolutionary history can be found in this paper, where the authors provide a historical overview of the sometimes meandering pathway of the Type III-CRISPR research and show that the archaeal HRAMP signature protein, which heretofore has had no assigned function, is a degenerate relative of Type-III-Cas signature protein Cas10, suggesting that HRAMP systems have descended from either their ancestors.
Journal ArticleDOI
PpCas9 from Pasteurella pneumotropica - a compact Type II-C Cas9 ortholog active in human cells.

O. Artamonova,Mikhail Khodorkovskii,Konstantin Severinov,Konstantin Severinov +24 more
TL;DR: Two Cas9 II-C orthologs, Df Cas9 from Defluviimonas sp.20V17 and PpCas9 from Pasteurella pneumotropica, are characterized and potentially can broaden the range of Cas9 applications in biomedicine and biotechnology.
