Journal ArticleDOI
Exploration of essential gene functions via titratable promoter alleles
Sanie Mnaimneh,Armaity P. Davierwala,Jennifer Haynes,Jason Moffat,Wen-Tao Peng,Wen Zhang,Xueqi Yang,Jeff Pootoolal,Gordon Chua,Andres Lopez,Miles Trochesset,Darcy L Morse,Nevan J. Krogan,Shawna L. Hiley,Zhijian Li,Quaid Morris,Jörg Grigull,Nicholas Mitsakakis,Christopher J. Roberts,Jack Greenblatt,Charles Boone,Chris A. Kaiser,Brenda J. Andrews,Timothy P. Hughes +23 more
TL;DR: This study created promoter-shutoff strains for over two-thirds of all essential yeast genes and subjected them to morphological analysis, size profiling, drug sensitivity screening, and microarray expression profiling, which identified genes involved in ribosome biogenesis, protein secretion, mitochondrial import, and tRNA charging.
Journal ArticleDOI
Regulation of chromosome stability by the histone H2A variant Htz1, the Swr1 chromatin remodeling complex, and the histone acetyltransferase NuA4.
Nevan J. Krogan,Kristin Baetz,Michael-Christopher Keogh,Nira Datta,Chika Sawa,Trevor C. Y. Kwok,Natalie J. Thompson,Michael Davey,Jeff Pootoolal,Timothy P. Hughes,Andrew Emili,Stephen Buratowski,Philip Hieter,Jack Greenblatt +13 more
TL;DR: NuA4, the only essential histone acetyltransferase complex in Saccharomyces cerevisiae, acetylates the N-terminal tails of histones H4 and H2A.
Journal ArticleDOI
Organization and function of APT, a subcomplex of the yeast cleavage and polyadenylation factor involved in the formation of mRNA and small nucleolar RNA 3'-ends.
Eduard Nedea,Xiaoyuan He,Minkyu Kim,Jeff Pootoolal,Guoqing Zhong,Veronica Canadien,Timothy P. Hughes,Stephen Buratowski,Claire Moore,Jack Greenblatt +9 more
TL;DR: A multiprotein complex, holo-CPF, containing the yeast cleavage and polyadenylation factor (CPF) and six additional polypeptides was isolated and use of mutant components of APT revealed that Ssu72 is important for preventing readthrough-dependent expression of downstream genes for both snoRNAs andpolyadenylated transcripts.
Journal ArticleDOI
Glycopeptide Antibiotic Resistance
TL;DR: Overcoming glycopeptide resistance will require innovative approaches to generate new antibiotics or otherwise to inhibit the action of resistance elements in various bacteria.
Journal ArticleDOI
Assembling the glycopeptide antibiotic scaffold: The biosynthesis of A47934 from Streptomyces toyocaensis NRRL15009.
Jeff Pootoolal,Michael G. Thomas,C. Gary Marshall,John M. Neu,Brian K. Hubbard,Christopher T. Walsh,Gerard D. Wright +6 more
TL;DR: The sequencing and annotation of the biosynthetic gene cluster for the glycopeptide antibiotic A47934 from Streptomyces toyocaensis NRRL15009 is reported, the first complete sequence for a teicoplanin class glycopePTide.
