TL;DR: An 800-million-member DNA-encoded library in which small molecules are covalently attached to an encoding oligonucleotide is reported, assembled by a combination of chemical and enzymatic synthesis, and interrogated by affinity selection.
Journal ArticleDOI
Carbamate analogues of fumagillin as potent, targeted inhibitors of methionine aminopeptidase-2.
Christopher C. Arico-Muendel,Dennis Benjamin,Teresa M Caiazzo,Paolo A. Centrella,Brooke D. Contonio,Charles M. Cook,Elisabeth Doyle,Gerhard Hannig,Matthew T. Labenski,Lily Lee Searle,Kenneth E Lind,Barry A. Morgan,Gary M. Olson,Christopher L. Paradise,Christopher Self,Steven R. Skinner,Barbara C. Sluboski,Jennifer L Svendsen,Charles D. Thompson,William F. Westlin,Kerry White +20 more
TL;DR: The synthesis and activity of fumagillin analogues that address the pharmacokinetic and safety liabilities of earlier candidates in this compound class are described and an improved pharmacokinetics profile relative to the earlier clinical candidate TNP-470 is demonstrated.
Journal ArticleDOI
Antiparasitic activities of novel, orally available fumagillin analogs.
Christopher C. Arico-Muendel,Paolo A. Centrella,Brooke D. Contonio,Barry A. Morgan,Gary K. O’Donovan,Christopher L. Paradise,Steven R. Skinner,Barbara Sluboski,Jennifer L Svendsen,Kerry White,Anjan Debnath,Jiri Gut,Nathan Wilson,James H. McKerrow,Joseph L. DeRisi,Philip J. Rosenthal,Peter K. Chiang +16 more
TL;DR: Activity of fumagillin analogs with an improved pharmacokinetic profile against malaria parasites, trypanosomes, and amoebas is demonstrated and forms a basis for further optimization of fomagillin based inhibitors against parasitic targets by inhibition of MetAP2.
Journal ArticleDOI

TL;DR: In the version of this article initially published, the IC50 values in the table in Figure 3c were listed as nM instead of μM, and the error has been corrected in the HTML and PDF versions of the article.
