Reactive oxygen species (ROS) are produced by living cells as normal cellular metabolic byproduct. Under excessive stress conditions, cells will produce numerous ROS, and the living organisms eventually evolve series of response mechanisms to adapt to the ROS exposure as well as utilize it as the signaling molecules. ROS molecules would trigger oxidative stress in a feedback mechanism involving many biological processes, such as apoptosis, necrosis and autophagy. Growing evidences have suggested that ROS play a critical role as the signaling molecules throughout the entire cell death pathway. Overwhelming production of ROS can destroy organelles structure and bio-molecules, which lead to inflammatory response that is a known underpinning mechanism for the development of diabetes and cancer. Cytochrome P450 enzymes (CYP) are regarded as the markers of oxidative stress, can transform toxic metabolites into ROS, such as superoxide anion, hydrogen peroxide and hydroxyl radical which might cause injury of cells. Accordingly, cells have evolved a balanced system to neutralize the extra ROS, namely antioxidant systems that consist of enzymatic antioxidants such as superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidases (GPxs), thioredoxin (Trx) as well as the non-enzymatic antioxidants which collectively reduce oxidative state. Herein, we review the recent novel findings of cellular processes induced by ROS, and summarize the roles of cellular endogenous antioxidant systems as well as natural anti-oxidative compounds in several human diseases caused by ROS in order to illustrate the vital role of antioxidants in prevention against oxidative stress.

Butyrate: A Double-Edged Sword for Health?

Branched chain amino acids (BCAAs), including leucine (Leu), isoleucine (Ile), and valine (Val), play critical roles in the regulation of energy homeostasis, nutrition metabolism, gut health, immunity and disease in humans and animals. As the most abundant of essential amino acids (EAAs), BCAAs are not only the substrates for synthesis of nitrogenous compounds, they also serve as signaling molecules regulating metabolism of glucose, lipid, and protein synthesis, intestinal health, and immunity via special signaling network, especially phosphoinositide 3-kinase/protein kinase B/mammalian target of rapamycin (PI3K/AKT/mTOR) signal pathway. Current evidence supports BCAAs and their derivatives as the potential biomarkers of diseases such as insulin resistance (IR), type 2 diabetes mellitus (T2DM), cancer, and cardiovascular diseases (CVDs). These diseases are closely associated with catabolism and balance of BCAAs. Hence, optimizing dietary BCAA levels should have a positive effect on the parameters associated with health and diseases. This review focuses on recent findings of BCAAs in metabolic pathways and regulation, and underlying the relationship of BCAAs to related disease processes.

A major challenge in affluent societies is the increase in disorders related to gut and metabolic health. Chronic over nutrition by unhealthy foods high in energy, fat, and sugar, and low in dietary fibre is a key environmental factor responsible for this development, which may cause local and systemic inflammation. A low intake of dietary fibre is a limiting factor for maintaining a viable and diverse microbiota and production of short-chain fatty acids in the gut. A suppressed production of butyrate is crucial, as this short-chain fatty acid (SCFA) can play a key role not only in colonic health and function but also at the systemic level. At both sites, the mode of action is through mediation of signalling pathways involving nuclear NF-κB and inhibition of histone deacetylase. The intake and composition of dietary fibre modulate production of butyrate in the large intestine. While butyrate production is easily adjustable it is more variable how it influences gut barrier function and inflammatory markers in the gut and periphery. The effect of butyrate seems generally to be more consistent and positive on inflammatory markers related to the gut than on inflammatory markers in the peripheral tissue. This discrepancy may be explained by differences in butyrate concentrations in the gut compared with the much lower concentration at more remote sites.

Impact of Diet-Modulated Butyrate Production on Intestinal Barrier Function and Inflammation.

Postbiotics are functional bioactive compounds, generated in a matrix during fermentation, which may be used to promote health. The term postbiotics can be regarded as an umbrella term for all synonyms and related terms of these microbial fermentation components. Therefore, postbiotics can include many different constituents including metabolites, short-chain fatty acids (SCFAs), microbial cell fractions, functional proteins, extracellular polysaccharides (EPS), cell lysates, teichoic acid, peptidoglycan-derived muropeptides and pili-type structures. Postbiotics is also a rather new term in the '-biotics' field. Where consensus exists for the definitions of pre- and probiotics, this is not yet the case for postbiotics. Here we propose a working definition and review currently known postbiotic compounds, their proposed mechanisms, clinical evidence and potential applications. Research to date indicates that postbiotics can have direct immunomodulatory and clinically relevant effects and evidence can be found for the use of postbiotics in healthy individuals to improve overall health and to relief symptoms in a range of diseases such as infant colic and in adults atopic dermatitis and different causes of diarrhea.

Postbiotics and Their Potential Applications in Early Life Nutrition and Beyond.

Antibiotics are one of the most important discoveries in the 20th century and have been widely used for treating animal diseases in the 21st century. However, antibiotic resistance among bacterial pathogens and widespread concerns regarding their use in animals has received great attention all over the world. Great attention has focused on scientific breakthroughs of the alternatives to antibiotics. Various materials such as enzymes, prebiotics, probiotics, minerals, antimicrobial peptides, acidifiers, plants and plant extracts have been tested as possible antibiotics alternatives. Owing to their effects on intestinal microbiota and immune function, research efforts have been conducted on the application of these feed supplements. This review highlights promising research results about the alternatives to antibiotics in animal husbandry that are expected to beneficially limit the adverse effects of antibiotics and ensure the safety of animal-derived foods and the environment.

The Signal Pathway of Antibiotic Alternatives on Intestinal Microbiota and Immune Function

Fasting is a simple metabolic strategy that is used to estimate the maintenance energy requirement where the energy supply for basic physiological functions is provided by the mobilization of body reserves. However, the underlying metabolic components of maintenance energy expenditure are not clear. This study investigated the differences in heat production (HP), respiratory quotient (RQ) and plasma metabolites in pigs in the fed and fasted state, using the techniques of indirect calorimetry and metabolomics. Nine barrows (45.2 ± 1.7 kg BW) were fed corn-soybean based meal diets and were kept in metabolism crates for a period of 14 d. After 7 d adaptation, pigs were transferred to respiratory chambers to determine HP and RQ based on indirect calorimetry. Pigs were fed the diet at 2,400 kJ ME/(kg BW0.6·d) during d 8 to 12. The last 2 d were divided into 24 h fasting and 48 h fasting treatment, respectively. Plasma samples of each pig were collected from the anterior vena cava during the last 3 d (1 d while pigs were fed and 2 d during which they were fasted). The metabolites of plasma were determined by high-resolution mass spectrometry using a metabolomics approach. Indirect calorimetry analysis revealed that HP and RQ were no significant difference between 24 h fasting and 48 h fasting, which were lower than those of fed state (P <  0.01). The nitrogen concentration of urine tended to decrease with fasting (P = 0.054). Metabolomics analysis between the fed and fasted state revealed differences in 15 compounds, most of which were not significantly different between 24 h fasting and 48 h fasting. Identified compounds were enriched in metabolic pathways related to linoleic acid metabolism, amino acid metabolism, sphingolipid metabolism, and pantothenate and CoA biosynthesis. These results suggest that the decreases in HP and RQ of growing pigs under fasting conditions were associated with the alterations of linoleic acid metabolism and amino acid metabolism. The integrative analysis also revealed that growing pigs under a 24-h fasting were more appropriate than a 48-h fasting to investigate the metabolic components of maintenance energy expenditure.

Simple Summary It has been well documented that the energy intake and associated energy retention of pigs will decline as the result of heat stress when the ambient temperature increases beyond thermoneutrality. The development of nutritional interventions to alleviate the negative effects of heat stress could be beneficial to pork producers. Studies have shown that reduced protein diets decrease the energy loss related to the excretion of excess N in urine and the energy loss associated with heat production. In conclusion, we provided a new nutritional strategy of decreasing dietary crude protein percentage from 12% to 10% in finishing pigs in summer, which had no negative effects on growth performance and carcass quality. Abstract The objective of this experiment was to investigate the effects of reduced dietary protein at natural high temperature in summer on the growth performance and carcass quality of finishing pigs. A total of 72 crossbreed pigs (Duroc × Landrace × Yorkshire) at an average body weight (BW) of 77 ± 5.7 kg were randomly assigned to two treatments, based on BW and sex, in six replicates per treatment, with six pigs per pen, using a randomized complete block design. The dietary crude protein (CP) level of the normal protein diet (NP) and the reduced protein diet (LP) were 12% and 10%, respectively. The growth performance and serum biochemical parameters of the pigs were analyzed for a 28-day experimental period. At the end of the experiment, 12 pigs were harvested to measure carcass characteristics and pork quality. The average highest ambient temperature during the experiment period was about 32.4 °C. There was a trend for the average daily feed intake (ADFI) to be lower in the pigs on the reduced protein diet compared to the control (p < 0.10) in the 0–28 day period. The serum urea nitrogen was lower (p < 0.05) for pigs fed the reduced protein diets only on day 14. The carcass characteristics and pork quality were not affected by dietary treatments. In conclusion, decreasing dietary crude protein percentage from 12% to 10% in finishing pigs in summer may have no negative effects on growth performance and carcass quality.
