Population Pharmacokinetics of Intravenous Polymyxin B in Critically Ill Patients: Implications for Selection of Dosage Regimens
Ana Maria Sandri,Cornelia B. Landersdorfer,Cornelia B. Landersdorfer,Jovan Jacob,Márcio Manozzo Boniatti,Micheline G. Dalarosa,Diego R. Falci,Tainá F. Behle,Rosaura C Bordinhao,Jiping Wang,Alan Forrest,Roger L. Nation,Jian Li,Alexandre P. Zavascki +13 more
TL;DR: This study showed that doses of intravenous polymyxin B are best scaled by total body weight, and dosage selection of this drug should not be based on renal function.
Journal ArticleDOI
New pharmacokinetic/pharmacodynamic studies of systemically administered colistin against Pseudomonas aeruginosa and Acinetobacter baumannii in mouse thigh and lung infection models: smaller response in lung infection
TL;DR: Colistin was substantially less effective in lung infection and the pharmacokinetic/pharmacodynamic target values will assist in the design of optimized dosage regimens.
Journal ArticleDOI
Antimicrobial Activity and Toxicity of the Major Lipopeptide Components of Polymyxin B and Colistin: Last-line Antibiotics against Multidrug-Resistant Gram-negative Bacteria.
Kade D. Roberts,Mohammad Abul Kalam Azad,Jiping Wang,Andrew S. Horne,Philip E. Thompson,Roger L. Nation,Tony Velkov,Jian Li +7 more
TL;DR: The results highlight the need to re-assess pharmacopoeial standards for polymyxins B and colistin and to standardize the composition of the different commercial products ofpolymyxin antibiotics.
Journal ArticleDOI
Teaching ‘Old’ Polymyxins New Tricks: New-Generation Lipopeptides Targeting Gram-Negative ‘Superbugs’
TL;DR: This report details the structure–activity relationships (SAR) based design, in toto synthesis, and preclinical evaluation of a series of novel polymyxin lipopeptides with better antibacterial activity against poly myxin-resistant Gram-negative bacteria.
Journal ArticleDOI
Pharmacokinetics/pharmacodynamics of systemically administered polymyxin B against Klebsiella pneumoniae in mouse thigh and lung infection models.
Cornelia B. Landersdorfer,Cornelia B. Landersdorfer,Jiping Wang,Veronika Wirth,Ke Chen,Keith S Kaye,Brian T. Tsuji,Jian Li,Jian Li,Roger L. Nation +9 more
TL;DR: Antibacterial activities of polymyxin B and colistin against thigh infection were compared at equimolar doses generating exposures resulting in maximal antibacterial activity and it was not possible to achieve stasis in lung infection, even at the highest dose tolerated by mice.
