A Selective Small Molecule Inhibitor of c-Met Kinase Inhibits c-Met-Dependent Phenotypes in Vitro and Exhibits Cytoreductive Antitumor Activity in Vivo
James G. Christensen,Randall E. Schreck,Jon Burrows,Poonam Kuruganti,Emily Chan,Phuong Le,Jeffrey H. Chen,Xueyan Wang,Lany Ruslim,Robert A. Blake,Kenneth E. Lipson,John Ramphal,Steven Do,Jingrong Jean Cui,Julie M. Cherrington,Dirk B. Mendel +15 more
TL;DR: The feasibility of selectively targeting c-Met with ATP-competitive small-molecules with high selectivity is demonstrated and the therapeutic potential of targetingc-Met in human cancers is suggested.
Journal ArticleDOI
The Met kinase inhibitor SU11274 exhibits a selective inhibition pattern toward different receptor mutated variants.
Sylvie Berthou,Daniel M. Aebersold,Laura S. Schmidt,Deborah Stroka,Christine Heigl,Bruno Streit,Denise Stalder,Guenther Gruber,Congxin Liang,Anthony R. Howlett,Daniel Candinas,Richard H. Greiner,Kenneth E. Lipson,Yitzhak Zimmer +13 more
TL;DR: It is reported that the Met kinase inhibitor SU11274 differentially affects the kinase activity and subsequent signaling of various mutant forms of Met, and the basis for the sensitivity or resistance to SU 11274 is discussed in terms of the position of the mutations predicted from a homology model.
Journal ArticleDOI
Transcriptional and posttranscriptional regulation of the proliferating cell nuclear antigen gene.
TL;DR: Results suggest a posttranscriptional component in the regulation of PCNA mRNA levels after serum stimulation but a transcriptional regulation by intron 4.
Journal ArticleDOI
Transcriptional activity of the human thymidine kinase gene determined by a method using the polymerase chain reaction and an intron-specific probe.
Kenneth E. Lipson,Renato Baserga +1 more
TL;DR: No TK hnRNA can be detected in truly quiescent human diploid fibroblasts, suggesting that in these cells, the TK gene is not transcribed in G0.
Journal Article
Potent and selective inhibitors of the Met [hepatocyte growth factor/scatter factor (HGF/SF) receptor] tyrosine kinase block HGF/SF-induced tumor cell growth and invasion.
Xueyan Wang,Phuong Le,Congxin Liang,Julie Chan,David Kiewlich,Todd W. Miller,Dave Harris,Li Sun,Audie Rice,Stefan Vasile,Robert A. Blake,Anthony R. Howlett,Neela Patel,Gerald Mcmahon,Kenneth E. Lipson +14 more
TL;DR: In this article, the pyrrole-indolinone inhibitors of Met tyrosine kinase have been found to inhibit HGF/SF-induced receptor phosphorylation in a dose-dependent manner.
