Journal ArticleDOI
Caspase-8 is the molecular switch for apoptosis, necroptosis and pyroptosis
Melanie Fritsch,Saskia Diana Günther,Robin Schwarzer,Marie-Christine Albert,Fabian Schorn,J. Paul Werthenbach,Lars Schiffmann,Neil Richard Stair,Hannah Stocks,Jens M. Seeger,Mohamed Lamkanfi,Martin Krönke,Manolis Pasparakis,Hamid Kashkar +13 more
TL;DR: It is shown that the expression of enzymatically inactive CASP8(C362S) causes embryonic lethality in mice by inducing necroptosis and pyroptosis, and prevents tissue damage during embryonic development and adulthood in mice.
Journal ArticleDOI
Embelin inhibits endothelial mitochondrial respiration and impairs neoangiogenesis during tumor growth and wound healing
Oliver Coutelle,Hue-Tran Hornig-Do,Axel Witt,Maria Andree,Lars Schiffmann,Michael Piekarek,Kerstin Brinkmann,Jens M. Seeger,Maxim Liwschitz,Satomi Miwa,Michael Hallek,Martin Krönke,Aleksandra Trifunovic,Sabine A. Eming,Rudolf J. Wiesner,Ulrich Hacker,Hamid Kashkar +16 more
TL;DR: It is shown that proliferating ECs increasingly depend on mitochondrial oxidative phosphorylation (OxPhos) for their increased energy demand, and the benzoquinone embelin is a weak mitochondrial uncoupler that prevents neoangiogenesis during tumor growth and wound healing by exhausting the low respiratory reserve of proliferatingECs without adversely affecting quiescent ECs.
Journal ArticleDOI
HER2/neu (ERBB2) expression and gene amplification correlates with better survival in esophageal adenocarcinoma.
Patrick Sven Plum,Florian Gebauer,Max Krämer,Hakan Alakus,Felix Berlth,Seung-Hun Chon,Lars Schiffmann,Thomas Zander,Reinhard Büttner,Arnulf H. Hölscher,Christiane Bruns,Alexander Quaas,Heike Loeser +12 more
TL;DR: A positive prognostic impact of HER2 overexpression was demonstrated in a large cohort of EAC, contrary to other solid malignancies including gastric cancer and breast cancer, but consistent to the results of a large study on EAC from 2012.
Journal ArticleDOI
Tumour-infiltrating neutrophils counteract anti-VEGF therapy in metastatic colorectal cancer
Lars Schiffmann,Melanie Fritsch,Florian Gebauer,Saskia Diana Günther,Neil Richard Stair,Jens M. Seeger,Fabinshy Thangarajah,Georg Dieplinger,Marc Bludau,Hakan Alakus,Heike Göbel,Alexander Quaas,Thomas Zander,Frank Hilberg,Christiane J. Bruns,Hamid Kashkar,Oliver Coutelle +16 more
TL;DR: The findings implicate CD177 as a biomarker for bevacizumab and suggest VEGF/Ang2 inhibition as a strategy to overcome neutrophil associated resistance to anti-angiogenic treatment.
Journal ArticleDOI
Evaluation of Angiopoietin-2 as a biomarker in gastric cancer: results from the randomised phase III AVAGAST trial
Ulrich Hacker,Laura Escalona-Espinosa,Nicola Consalvo,Valentin Goede,Lars Schiffmann,Stefan Scherer,Priti Hedge,Eric Van Cutsem,Oliver Coutelle,Hildegard Büning,Hildegard Büning +10 more
TL;DR: Angiopoietin-2 (Ang-2), a key driver of tumour angiogenesis, metastasis and resistance to antiangiogenic treatment, is studied as a biomarker for OS in advanced gastric cancer strongly associated with LM.
