The Hitchhiker's Guide to Flow Chemistry

We present a droplet-based microfluidic technology that enables high-throughput screening of single mammalian cells. This integrated platform allows for the encapsulation of single cells and reagents in independent aqueous microdroplets (1 pL to 10 nL volumes) dispersed in an immiscible carrier oil and enables the digital manipulation of these reactors at a very high-throughput. Here, we validate a full droplet screening workflow by conducting a droplet-based cytotoxicity screen. To perform this screen, we first developed a droplet viability assay that permits the quantitative scoring of cell viability and growth within intact droplets. Next, we demonstrated the high viability of encapsulated human monocytic U937 cells over a period of 4 days. Finally, we developed an optically-coded droplet library enabling the identification of the droplets composition during the assay read-out. Using the integrated droplet technology, we screened a drug library for its cytotoxic effect against U937 cells. Taken together our droplet microfluidic platform is modular, robust, uses no moving parts, and has a wide range of potential applications including high-throughput single-cell analyses, combinatorial screening, and facilitating small sample analyses.

Based on the principles and metrics of green chemistry and sustainable development, biocatalysis is both a green and sustainable technology. This is largely a result of the spectacular advances in molecular biology and biotechnology achieved in the past two decades. Protein engineering has enabled the optimization of existing enzymes and the invention of entirely new biocatalytic reactions that were previously unknown in Nature. It is now eminently feasible to develop enzymatic transformations to fit predefined parameters, resulting in processes that are truly sustainable by design. This approach has successfully been applied, for example, in the industrial synthesis of active pharmaceutical ingredients. Role of Biocatalysis in Sustainable Chemistry

Continuous-flow photochemistry in microreactors receives a lot of attention from researchers in academia and industry as this technology provides reduced reaction times, higher selectivities, straightforward scalability, and the possibility to safely use hazardous intermediates and gaseous reactants. In this review, an up-to-date overview is given of photochemical transformations in continuous-flow reactors, including applications in organic synthesis, material science, and water treatment. In addition, the advantages of continuous-flow photochemistry are pointed out and a thorough comparison with batch processing is presented.

Applications of Continuous-Flow Photochemistry in Organic Synthesis, Material Science, and Water Treatment

Biodiesel can be produced from vegetable oils, animal fats, and waste cooking oils by transesterification with ethanol (also called ethanolysis) in order to substitute fossil fuels. In this work, we were interested in the transesterification reaction of sunflower oil with ethanol, which leads to ethyl esters, used to date for applications principally in food and cosmetic industry. To open the application field to biofuels (to substitute current fuels resulting from fossil resources), the process efficiency has to be developed to be economically profitable. The batch reaction of vegetable oil ethanolysis was transposed to a micro-scaled continuous device (PFA tube of 508 μm internal diameter), inducing better heat and mass transfer. Study of the influence of the operational conditions (reactants flow, initial ethanol to oil molar ratio, temperature. . .) revealed the favourable reaction parameters necessary to reach high conversions and yields. In these conditions, it is possible to acquire kinetics data at the first seconds of the reaction, which was not feasible in a conventional batch process. These data were used to model occurring phenomena and to determine kinetic constants and transfer coefficients. The model was subsequently used to simulate reactions with other operational conditions. To acquire these data in microreactors, an on-line analysis method by Near InfraRed (NIR) spectroscopy was developed by using gas chromatography as a reference method. PLS models were then set up to quantify on-line the major compounds contents during the reaction.

New internals have been designed and implemented in a semi-industrial continuous solid-liquid pulsed column. By coupling these internals with a non-sinusoidal pulsation, a better control of polydispersed solid particles is achieved. The pulsation is composed of a mixing step during which only the liquid flows through the column and an impulsion step designed to transport the solid phase from stage to stage. Solid and liquid phase behaviors are characterized thanks to residence time distribution measurements. This study demonstrates the strong impact of the pulsation and of the liquid flow rate during the impulsion in reducing particle segregation inside the pilot. The effects of operating parameters on the liquid phase are also investigated and the choice of an operating compromise is discussed to balance the advantages and drawbacks of the process.

Use of Pulsation to Control Polydispersed Particle Flow in a New Type of Pulsed Column

Implementation of exothermic and fast reactions requires a perfect temperature control to avoid thermal runaway and in most cases to limit by-products production. In order to fit with the heat removal capacity of common devices, expensive strategies are currently used to slow down this kind of reactions in order to avoid a strong temperature increase such as reactants dilution. Within the concept of process intensification, industries could move towards more sustainable process by reducing technology constraints to the benefit of chemistry. For that purpose, a two-step methodology is implemented. The first step consists in the optimisation of the operating conditions only based on stoichio-chemical scheme and kinetic laws. This is carried out by adjusting temperature profile and feeding rate strategy in a batch operation. Then a design for a continuous process is proposed, trying to approach the optimal batch operating conditions. This methodology is applied to the linear alkylbenzene sulfonation.
