Journal ArticleDOI
The mitochondrial kinase PINK1: functions beyond mitophagy.
TL;DR: Mounting evidence indicates that PINK1 has additional functions, for example, in regulating complex I activity and maintaining neuronal viability in response to stress.
Journal ArticleDOI
A protein quality control pathway regulated by linear ubiquitination.
Eva M van Well,Verian Bader,Maria Patra,Ana Sánchez-Vicente,Jens Meschede,Nikolas Furthmann,Cathrin Schnack,Alina Blusch,Joseph Longworth,Elisabeth Petrasch-Parwez,Kohji Mori,Thomas Arzberger,Thomas Arzberger,Dietrich Trümbach,Lena Angersbach,Cathrin Showkat,Dominik A. TL;DR: This work reports that various disease‐associated protein aggregates are modified by the linear ubiquitin chain assembly complex (LUBAC), identifying linear Ubiquitination as a protein quality control mechanism and hence a novel target for disease‐modifying strategies in proteinopathies.
Journal ArticleDOI
The parkin-coregulated gene product PACRG promotes TNF signaling by stabilizing LUBAC
Jens Meschede,Maria Šadić,Nikolas Furthmann,Tim Miedema,Dominik A. Sehr,A. Kathrin Müller-Rischart,Verian Bader,Lena A. Berlemann,Anna Pilsl,Anita Schlierf,Katalin Barkovits,Barbara Kachholz,Katrin Rittinger,Fumiyo Ikeda,Katrin Marcus,Liliana Schaefer,Jörg Tatzelt,Jörg Tatzelt,Konstanze F. Winklhofer,Konstanze F. Winklhofer +19 more
TL;DR: It is found that PACRG played no role in mitophagy in cultured human cells but did promote NF-κB signaling downstream of the tumor necrosis factor (TNF) receptor TNFR1 and protected cells from TNF-induced apoptosis by interacting with and stabilizing the linear ubiquitin chain assembly complex (LUBAC).
Journal ArticleDOI
LUBAC assembles a ubiquitin signaling platform at mitochondria for signal amplification and transport of NF‐κB to the nucleus
Zhixiao Wu,Lena A. Berlemann,Verian Bader,Dominik A. Sehr,Eva Dawin,Alberto Covallero,Jens Meschede,Lena Angersbach,Cathrin Showkat,Jonas B. Michaelis,Christian Münch,Bettina Rieger,Dmitry Namgaladze,M.G. Herrera,Fabienne C. Fiesel,Wolfdieter Springer,Marta Mendes,Jennifer Stepien,Katalin Barkovits,Katrin Marcus,Albert Sickmann,Gunnar Dittmar,Karin B. Busch,Dietmar Riedel,Marisa Brini,Jörg Tatzelt,Tito Calì,Konstanze F. Winklhofer +27 more
TL;DR: In this paper , the authors show that the NF-κB pathway activated by TNF employs mitochondria as a platform for signal amplification and shuttling of activated NF•κB to the nucleus.
Journal ArticleDOI
Parkin — eine neuroprotektive E3-Ubiquitin-Ligase
TL;DR: The prominent role of mitochondria in energy supply and cellular homeostasis highlights the importance of mitochondrial quality control mechanisms to preserve organelle function under stress and to eliminate irreversibly damaged mitochondria.
