MicroRNA-21 in Pancreatic Cancer: Correlation with Clinical Outcome and Pharmacologic Aspects Underlying Its Role in the Modulation of Gemcitabine Activity
Elisa Giovannetti,Niccola Funel,Godefridus J. Peters,Marco Del Chiaro,Leyla A. Erozenci,Enrico Vasile,Leticia G. Leon,Luca Pollina,Annemieke Groen,Alfredo Falcone,Romano Danesi,Daniela Campani,Henk M.W. Verheul,Ugo Boggi +13 more
TL;DR: Modulation of apoptosis, Akt phosphorylation, and expression of genes involved in invasive behavior may contribute to the role of miR-21 in gemcitabine chemoresistance and to the rational development of new targeted combinations.
Journal ArticleDOI
Identification of microRNA-21 as a biomarker for chemoresistance and clinical outcome following adjuvant therapy in resectable pancreatic cancer.
Jin Hyeok Hwang,Jin Hyeok Hwang,Jin Hyeok Hwang,Johannes Voortman,Johannes Voortman,Elisa Giovannetti,Elisa Giovannetti,Seth M. Steinberg,Leticia G. Leon,Yong-Tae Kim,Niccola Funel,Joo Kyung Park,Min A Kim,Gyeong Hoon Kang,Sun-Whe Kim,Marco Del Chiaro,Godefridus J. Peters,Giuseppe Giaccone +17 more
TL;DR: Evidence is provided that miR-21 may allow stratification for adjuvant therapy, and represents a new potential target for therapy in PDAC, as well as the single most predictive biomarker for treatment outcome.
Journal ArticleDOI
Discovery of N-hydroxyindole-based inhibitors of human lactate dehydrogenase isoform A (LDH-A) as starvation agents against cancer cells.
Carlotta Granchi,Sarabindu Roy,Chiara Giacomelli,Marco Macchia,Tiziano Tuccinardi,Adriano Martinelli,Mario Lanza,Laura Betti,Gino Giannaccini,Antonio Lucacchini,Nicola Funel,Leticia G. Leon,Leticia G. Leon,Elisa Giovannetti,Godefridus J. Peters,Rahul Palchaudhuri,Emilia C. Calvaresi,Paul J. Hergenrother,Filippo Minutolo +18 more
TL;DR: New and efficient N-hydroxyindole-based inhibitors of LDH-A are discovered, which are isoform-selective (overLDH-B) and competitive with both the substrate (pyruvate) and the cofactor (NADH) and they proved to be particularly effective under hypoxic conditions.
Journal ArticleDOI
Antiproliferative activity of synthetic naphthoquinones related to lapachol. First synthesis of 5-hydroxylapachol.
Evelyn L. Bonifazi,Carla Ríos-Luci,Leticia G. Leon,Gerardo Burton,José M. Padrón,Rosana I. Misico +5 more
TL;DR: A series of 5-hydroxy-1,4-naphthoquinones analogues was synthesized from juglone and their antiproliferative activity against a representative panel of six human solid tumor cell lines has been investigated and the results provide insight into the correlation between some structural properties of 4,5-dihydroxy-3-(3-methylbut-2-enyl)naphthalene-1-4-dione.
Journal ArticleDOI
High-Throughput MicroRNA (miRNAs) Arrays Unravel the Prognostic Role of MiR-211 in Pancreatic Cancer
Elisa Giovannetti,Arjan van der Velde,Niccola Funel,Enrico Vasile,Vittorio Perrone,Leticia G. Leon,Nelide De Lio,Amir Avan,S. Caponi,Luca Pollina,Valentina Galla,Hiroko Sudo,Alfredo Falcone,Daniela Campani,Ugo Boggi,Godefridus J. Peters +15 more
TL;DR: Through comprehensive microarray analysis and PCR validation, miR-211 emerged as the best discriminating miRNA, with significantly higher expression in long- vs. short-OS patients, and was identified as a prognostic factor in resected PDAC.
