Journal ArticleDOI
Structural basis for glucose tolerance in GH1 β-glucosidases
Priscila Oliveira de Giuseppe,Tatiana de Arruda Campos Brasil de Souza,Flavio Henrique Moreira Souza,Letícia Maria Zanphorlin,Carla Botelho Machado,Richard J. Ward,João Atílio Jorge,Rosa dos Prazeres Melo Furriel,Mário T. Murakami +8 more
TL;DR: The first crystal structure of a fungal β-glucosidase stimulated by glucose was solved in native and glucose-complexed forms, revealing that the shape and electrostatic properties of the entrance to the active site, including the +2 subsite, determine glucose tolerance.
Journal ArticleDOI
Purification and characterization of a new alkaline serine protease from the thermophilic fungus Myceliophthora sp.
Letícia Maria Zanphorlin,Hamilton Cabral,Eliane Candiani Arantes,Diego M. Assis,Luiz Juliano,Maria A. Juliano,R. Da-Silva,Eleni Gomes,Gustavo Orlando Bonilla-Rodriguez +8 more
TL;DR: The results showed that Abz-KLISSKQ-EDDnp is the best substrate among those tested for the purified protease (kcat/Km = 1275.3 mM −1 s −1 ).
Journal ArticleDOI
A novel cold-adapted and glucose-tolerant GH1 β-glucosidase from Exiguobacterium antarcticum B7.
Elaine Crespim,Letícia Maria Zanphorlin,Flavio Henrique Moreira Souza,José A. Diogo,Alex C. Gazolla,Carla Botelho Machado,Fernanda Lopes de Figueiredo,Amanda Silva de Sousa,Felipe Resende Nóbrega,Vivian Helena Pellizari,Mário T. Murakami,Roberto Ruller +11 more
TL;DR: A novel GH1 β-glucosidase from a bacterium isolated from Antarctica soil samples was recombinantly overexpressed in Escherichia coli cells and characterized, showing unusual pH dependence and high tolerance for product inhibition.
Journal ArticleDOI
Oligomerization as a strategy for cold adaptation: Structure and dynamics of the GH1 β-glucosidase from Exiguobacterium antarcticum B7
Letícia Maria Zanphorlin,Priscila Oliveira de Giuseppe,Rodrigo V. Honorato,Celisa Caldana Costa Tonoli,Juliana Fattori,Elaine Crespim,Paulo S. L. Oliveira,Roberto Ruller,Mário T. Murakami +8 more
TL;DR: This work used X-ray crystallography, in solution studies and molecular dynamics simulations to reveal the structural basis for cold adaptation of the GH1 β-glucosidase from Exiguobacterium antarcticum B7 and reveals a novel strategy for enzyme cold adaptation.
Journal ArticleDOI
Molecular mechanisms associated with xylan degradation by Xanthomonas plant pathogens
Camila R. Santos,Zaira B. Hoffmam,Vanesa Peixoto de Matos Martins,Letícia Maria Zanphorlin,Leandro H.P. Assis,Rodrigo V. Honorato,Paulo S. L. Oliveira,Roberto Ruller,Mário T. Murakami +8 more
TL;DR: Results indicate that XynB contributes to breach the plant cell wall barrier, providing nutrients and facilitating the translocation of effector molecules, whereas the exo-oligoxylanase XynA possibly participates in the suppression of oligosaccharide-induced immune responses.
