Journal ArticleDOI
Deciphering the targets of retroviral protease inhibitors in Plasmodium berghei.
Noah Machuki Onchieku,Reagan M. Mogire,Reagan M. Mogire,Loise Ndung'u,Peter Mwitari,Francis Kimani,Damaris Matoke-Muhia,Daniel Kiboi,Gabriel Magoma,Gabriel Magoma +9 more
TL;DR: It is concluded that PM4 plays a vital role in assuring asexual stage parasite fitness and might be mediating LP and SQ action.
Journal ArticleDOI
Amodiaquine resistance in Plasmodium berghei is associated with PbCRT His95Pro mutation, loss of chloroquine, artemisinin and primaquine sensitivity, and high transcript levels of key transporters.
Loise Ndung'u,Loise Ndung'u,Benard Langat,Esther Magiri,Joseph K. Ng’ang’a,Beatrice Irungu,Alexis Nzila,Daniel Kiboi,Daniel Kiboi,Daniel Kiboi +9 more
TL;DR: The selection of amodiaquine resistance yielded stable “multidrug-resistant’’ parasites and thus may be used to study shared resistance mechanisms associated with other antimalarial drugs.
Journal ArticleDOI
Amodiaquine Resistance in Plasmodium Berghei is Associated with Pbcrt His95Pro Mutation, Loss of Chloroquine, Artemisinin and Primaquine Sensitivity, and High Transcript Levels of Key Transporters [Version 2; Referees: 3 Approved]
Loise Ndung'u,Bernard Langat,E. Magiri,Joseph K. Ng’ang’a,Beatrice Irungu,Alexis Nzila,Daniel Kiboi +6 more
Posted ContentDOI
Efficacy of Lumefantrine against piperaquine resistant Plasmodium berghei parasites is selectively restored by probenecid, verapamil, and cyproheptadine through ferredoxin NADP+-reductase and cysteine desulfurase
Fagdéba David Bara,Loise Ndung'u,Noah Machuki Onchieku,Beatrice Irungu,Simplice Damintoti Karou,Francis Kimani,Damaris Matoke-Muhia,Peter Mwitari,Gabriel Magoma,Alexis Nzila,Daniel Kiboi +10 more
TL;DR: It is demonstrated that the incorporation of any of the RA into an antimalarial combination that comprises LM would augment LM activity and concomitantly antagonize the emergence of LM resistance derived from PQ pressure, and clues on the link between SUFS and FNR are provided.
Posted ContentDOI
Genetic alteration of ferredoxin NADP+-reductase or cysteine desulfurase in piperaquine-resistant Plasmodium berghei restores susceptibility to lumefantrine and abolishes the impact of probenecid, verapamil, and cyproheptadine
Fagdéba David Bara,Fagdéba David Bara,Loise Ndung'u,Noah Machuki Onchieku,Beatrice Irungu,Simplice Damintoti Karou,Francis Kimani,Damaris Matoke-Muhia,Peter Mwitari,Gabriel Magoma,Gabriel Magoma,Alexis Nzila,Daniel Kiboi +12 more
TL;DR: Deletion of the SufS or overexpression of the FNR genes in the PQR abolished the impact of the chemosensitizers on the LM activity, and restored the susceptibility of the P QR parasites to LM.
