Efficacy of NS5A Inhibitors Against Hepatitis C Virus Genotypes 1–7 and Escape Variants
Judith M. Gottwein,Long V. Pham,Lotte S. Mikkelsen,Lubna Ghanem,Santseharay Ramirez,Troels K. H. Scheel,Thomas H. R. Carlsen,Jens Bukh +7 more
TL;DR: A comprehensive comparison of the efficacy of the 7 clinically relevant inhibitors of HCV NS5A and identified variants associated with resistance to each agent are performed, which could improve treatment of patients with HCV infection.
Journal ArticleDOI
Overcoming culture restriction for SARS-CoV-2 in human cells facilitates the screening of compounds inhibiting viral replication.
Santseharay Ramirez,Santseharay Ramirez,Carlota Fernandez-Antunez,Carlota Fernandez-Antunez,Andrea Galli,Andrea Galli,Alexander Underwood,Alexander Underwood,Long V. Pham,Long V. Pham,Line Abildgaard Ryberg,Line Abildgaard Ryberg,Shan Feng,Shan Feng,Martin Schou Pedersen,Martin Schou Pedersen,Lotte S. Mikkelsen,Lotte S. Gottwein,Ulrik Fahnøe,Ulrik Fahnøe,Jens Bukh,Jens Bukh +31 more
TL;DR: In this paper, serial passages of a SARS-CoV-2 isolate (original-SARS2) in the human hepatoma cell clone Huh7.5 led to the selection of a variant (adapted SARS2), which significantly improved infectivity in human liver (Huh7 and Huh75) and lung cancer (unmodified Calu-1 and A549) cells.
Posted ContentDOI
In vitro efficacy of Artemisinin-based treatments against SARS-CoV-2
Kerry Gilmore,Yuyong Zhou,Santseharay Ramirez,Long V. Pham,Ulrik Fahnøe,Shan Feng,Anna Offersgaard,Jakob Trimpert,Jens Bukh,Klaus Osterrieder,Klaus Osterrieder,Judith M. Gottwein,Peter H. Seeberger,Peter H. Seeberger +13 more
TL;DR: In vitro efficacy of Artemisia annua extracts as well as artemisinin, artesunate, and artemether against SARS-CoV-2 is reported.
Journal ArticleDOI
Stearoyl Coenzyme A Desaturase 1 Is Associated with Hepatitis C Virus Replication Complex and Regulates Viral Replication
TL;DR: Manipulation of SCD1 activity may represent a novel host-targeted antiviral strategy for the treatment of HCV infection and provide novel mechanistic insights into the roles ofSCD1.
Journal ArticleDOI
HCV Genotype 6a Escape From and Resistance to Velpatasvir, Pibrentasvir, and Sofosbuvir in Robust Infectious Cell Culture Models.
Long V. Pham,Santseharay Ramirez,Judith M. Gottwein,Ulrik Fahnøe,Yi-Ping Li,Jannie Pedersen,Jens Bukh +6 more
TL;DR: Strains of HCV genotype 6a isolated from patients can be adapted to propagate in cultured cells, permitting studies of the complete life cycle for this important genotype, which quickly becomes resistant to single inhibitors via the rapid emergence and persistence of RAS.
