Characterization of SARS2 Nsp15 nuclease activity reveals it's mad about U.
Meredith N Frazier,Lucas B. Dillard,Juno M. Krahn,Lalith Perera,Jason Williams,Isha M Wilson,Zachary D. Stewart,Monica C. Pillon,Leesa J. Deterding,Mario J. Borgnia,Robin E. Stanley +10 more
TL;DR: In this paper, the authors used cryo-EM to capture structures of Nsp15 bound to RNA in pre-and post-cleavage states, and determined how the sequence of the RNA substrate dictates cleavage and found that outside of polyU tracts, nsp15 has a strong preference for purines 3' of the cleaved uridine.
Journal ArticleDOI
Structural basis of NPR1 in activating plant immunity
Shivesh Kumar,Raul Zavaliev,Qinglin Wu,Ye Zhou,Jie Cheng,Lucas B. Dillard,Jordan J. Powers,John Withers,Jinshi Zhao,Ziqiang Guan,Mario J. Borgnia,Alberto Bartesaghi,Xinnian Dong,Pei Zhou +13 more
TL;DR: In this paper , the structure of NPR1 and its complex with the transcription factor TGA3 was analyzed using Cryo-electron microscopy and crystal structure analysis, which revealed a unique zinc-finger motif in BTB for interacting with ankyrin repeats and mediating NPR1 oligomerization, providing a structural explanation for a direct role of salicylic acid in regulating NPR1dependent gene expression.

Journal ArticleDOI
Cryo-EM structures of the SARS-CoV-2 endoribonuclease Nsp15 reveal insight into nuclease specificity and dynamics.
Monica C. Pillon,Monica C. Pillon,Meredith N Frazier,Lucas B. Dillard,Jason Williams,Seda Kocaman,Juno M. Krahn,Lalith Perera,Cassandra K. Hayne,Jacob Gordon,Zachary D. Stewart,Mack Sobhany,Leesa J. Deterding,Allen L. Hsu,Venkata P. Dandey,Mario J. Borgnia,Robin E. Stanley +16 more
TL;DR: In this paper, a series of cryo-EM reconstructions of SARS-CoV-2 Nsp15, in both apo and UTP-bound states, are presented.
Journal ArticleDOI
Tau induces formation of α-synuclein filaments with distinct molecular conformations.
Alimohammad Hojjatian,Anvesh K. R. Dasari,Urmi Sengupta,Dianne W. Taylor,Nadia Daneshparvar,Fatemeh Abbasi Yeganeh,Lucas B. Dillard,Brian Michael,Robert G. Griffin,Mario J. Borgnia,Rakez Kayed,Kenneth A. Taylor,Kwang Hun Lim +12 more
TL;DR: In this paper, structural characterizations of α-synuclein filaments formed in the presence of a potential co-factor, tau, using cryo-EM and solid-state NMR were performed.
