Journal ArticleDOI
Discovery of the First C-Nucleoside HCV Polymerase Inhibitor (GS-6620) with Demonstrated Antiviral Response in HCV Infected Patients
A. Cho,Lijun Zhang,J. Xu,R. Lee,Thomas Butler,Sammy Metobo,Aktoudianakis,Willard Lew,Hong Ye,Michael O. Clarke,Edward Doerffler,Daniel Byun,Ting Wang,Darius Babusis,Anne Carey,P. German,D. Sauer,Weidong Zhong,S. Rossi,Martijn Fenaux,John G. McHutchison,Jason K. Perry,Joy Y. Feng,Adrian S. Ray,Cynthia Kim +24 more
TL;DR: The discovery of a C-nucleoside class of NS5B inhibitors of Hepatitis C virus is reported, with potential for potent activity but with high intra- and interpatient pharmacokinetic and pharmacodynamic variability.
Journal ArticleDOI
Extraintestinal spread and replication of a homologous EC rotavirus strain and a heterologous rhesus rotavirus in BALB/c mice.
TL;DR: Comparisons in newborn mice indicate that extraintestinal spread and replication occurs commonly during homologous and some heterologous rotaviral infections; that the substantial host range restrictions for rhesus rotavirus, aheterologous strain present in the intestine, are not necessarily apparent at systemic sites.
Journal ArticleDOI
Mechanistic Characterization of GS-9190 (Tegobuvir), a Novel Nonnucleoside Inhibitor of Hepatitis C Virus NS5B Polymerase
I-hung Shih,Inge Vliegen,Betty Peng,Huiling Yang,Christy M. Hebner,Jan Paeshuyse,Gerhard Pürstinger,Martijn Fenaux,Yang Tian,Eric Mabery,Xiaoping Qi,Gina Bahador,Matthew Paulson,Laura S. Lehman,Steven S. Bondy,Tse Winston C,Hans Reiser,William A. Lee,Uli Schmitz,Johan Neyts,Weidong Zhong +20 more
TL;DR: It is demonstrated that GS-9190 represents a novel class of nonnucleoside polymerase inhibitors that interact with NS5B likely through involvement of the β-hairpin in the thumb subdomain.
Journal ArticleDOI
Role of Interferon in Homologous and Heterologous Rotavirus Infection in the Intestines and Extraintestinal Organs of Suckling Mice
Ningguo Feng,Bumseok Kim,Martijn Fenaux,H. Nguyen,Phuoc T. Vo,M. B. Omary,Harry B. Greenberg +6 more
TL;DR: The data indicate that the requirements for the interferon system to inhibit intestinal and extraintestinal viral replication in suckling mice vary among different heterologous and homologous rotavirus strains, and this variation is associated with lethal systemic disease.
Journal ArticleDOI
Quantitative Evaluation of Rotaviral Antigenemia in Children with Acute Rotaviral Diarrhea
Pratima Ray,Martijn Fenaux,Sumit Sharma,Jyoti Malik,Swati Subodh,Shinjini Bhatnagar,Harry B. Greenberg,Roger I. Glass,Jon R. Gentsch,Maharaj K. Bhan +9 more
TL;DR: Serum levels of both antigen and RNA were inversely associated with baseline titers of rotaviral serum immunoglobulin G, andAntigenemia was associated with infection with G1 strains and with low baseline titer of rotviral serum antibody.
