TL;DR: It is shown that TRIP12 and UBR5, two HECT domain ubiquitin E3 ligases, control accumulation of RNF168, a rate-limiting component of a pathway that ubiquitylates histones after DNA breakage and can be saturated by increasing amounts of DSBs.

TL;DR: Results generally support PAR levels as potential candidate biomarker, but additional studies are needed to determine whether PAR detection is technically feasible in tissue biopsies, and whether PARP inhibitor sensitivity is inevitably associated with detectable amounts of PAR, especially in light of the limited sensitivity of the available antibodies for shorter PAR chains.
