TRIP12 and UBR5 Suppress Spreading of Chromatin Ubiquitylation at Damaged Chromosomes
Thorkell Gudjonsson,Matthias Altmeyer,Velibor Savic,Luis I. Toledo,Christoffel Dinant,Merete Grøfte,Jirina Bartkova,Maria Poulsen,Yasuyoshi Oka,Simon Bekker-Jensen,Niels Mailand,Beate Neumann,Jean-Karim Hériché,Robert F. Shearer,Darren N. Saunders,Jiri Bartek,Jiri Lukas,Claudia Lukas +17 more
TL;DR: It is shown that TRIP12 and UBR5, two HECT domain ubiquitin E3 ligases, control accumulation of RNF168, a rate-limiting component of a pathway that ubiquitylates histones after DNA breakage and can be saturated by increasing amounts of DSBs.
Journal ArticleDOI
Adipocyte lipolysis links obesity to breast cancer growth: adipocyte-derived fatty acids drive breast cancer cell proliferation and migration
Seher Balaban,Robert F. Shearer,Lisa S. Lee,Michelle van Geldermalsen,Mark Schreuder,Mark Schreuder,Harrison C. Shtein,Rose Cairns,Kristen C. Thomas,Daniel J. Fazakerley,Thomas Grewal,Jeff Holst,Darren N. Saunders,Darren N. Saunders,Andrew J. Hoy +14 more
TL;DR: It is shown that co-culture of breast cancer cells with adipocytes revealed cancer cell-stimulated depletion of adipocyte triacylglycerol, highlighting a novel and potentially important role for adipocyte lipolysis in the provision of metabolic substrates to Breast cancer cells, thereby supporting cancer progression.
Journal ArticleDOI
Tyrosine phosphorylation profiling reveals the signaling network characteristics of basal breast cancer cells
Falko Hochgräfe,Luxi Zhang,Sandra A O'Toole,Brigid C. Browne,Mark Pinese,Ana Porta Cubas,Gillian M. Lehrbach,David R. Croucher,Danny Rickwood,Alice Boulghourjian,Robert F. Shearer,Radhika Nair,Alexander Swarbrick,Dana Faratian,Peter Mullen,David J. Harrison,Andrew V. Biankin,Robert L. Sutherland,Mark J. Raftery,Roger J. Daly +19 more
TL;DR: It is indicated that patient stratification based on expression/activation of drug targets, coupled with use of multi-kinase inhibitors or combination therapies, may be required for effective treatment of this breast cancer subgroup.
Journal ArticleDOI
Hedgehog Overexpression Is Associated with Stromal Interactions and Predicts for Poor Outcome in Breast Cancer
Sandra A O'Toole,Dorothy A Machalek,Robert F. Shearer,Ewan K.A. Millar,Radhika Nair,Peter R. Schofield,Duncan J. Mcleod,Caroline L. Cooper,Catriona M. McNeil,Andrea McFarland,Akira Nguyen,Christopher J. Ormandy,Min Ru Qiu,Brian Rabinovich,Luciano G. Martelotto,Duc Vu,Gregory E. Hannigan,Elizabeth A. Musgrove,Daniel Christ,Robert L. Sutherland,D. Neil Watkins,Alexander Swarbrick +21 more
TL;DR: In a cohort of 279 patients with invasive ductal carcinoma of the breast, expression of Hh ligand was significantly associated with increased risk of metastasis, breast cancer-specific death, and a basal-like phenotype, and paracrine signature was an independent predictor for overall survival in multivariate analysis.
Journal ArticleDOI
Functional Roles of the E3 Ubiquitin Ligase UBR5 in Cancer.
TL;DR: Emerging molecular insights indicate a role for UBR5 in integrating/coordinating various cellular signaling pathways and highlight the need to systematically characterize substrates, and address limitations in current animal models, to better define the role of U BR5 in cancer.
