Journal ArticleDOI
Subtypes of pancreatic ductal adenocarcinoma and their differing responses to therapy.
Eric A. Collisson,Anguraj Sadanandam,Anguraj Sadanandam,Peter Olson,Peter Olson,William J. Gibb,William J. Gibb,Morgan L. Truitt,Shenda Gu,J. Cooc,Jennifer Weinkle,Grace E. Kim,Lakshmi Jakkula,Heidi S. Feiler,Andrew H. Ko,Adam B. Olshen,Kathleen L Danenberg,Margaret A. Tempero,Paul T. Spellman,Douglas Hanahan,Douglas Hanahan,Joe W. Gray,Joe W. Gray +22 more
TL;DR: Three PDA subtypes are defined: classical, quasimesenchymal and exocrine-like, and evidence for clinical outcome and therapeutic response differences between them is presented, and gene signatures for these subtypes that may have utility in stratifying patients for treatment are defined.
Journal ArticleDOI
Cancer-Associated Fibroblasts Are Activated in Incipient Neoplasia to Orchestrate Tumor-Promoting Inflammation in an NF-κB-Dependent Manner
TL;DR: It is shown that normal dermal fibroblasts can be "educated" by carcinoma cells to express proinflammatory genes, and this ability to "educate" them is shown to be related to tumor-enhancing inflammation.
Journal ArticleDOI
IRE1α Cleaves Select microRNAs During ER Stress to Derepress Translation of Proapoptotic Caspase-2
John-Paul Upton,Likun Wang,Dan Han,Eric S. Wang,Noelle E. Huskey,Lionel Lim,Morgan L. Truitt,Michael T. McManus,Davide Ruggero,Andrei Goga,Feroz R. Papa,Scott A. Oakes +11 more
TL;DR: It is reported that sustained IRE1α RNase activation caused rapid decay of select microRNAs that normally repress translation of Caspase-2 mRNA, and thus sharply elevates protein levels of this initiator protease of the mitochondrial apoptotic pathway.
Journal ArticleDOI
Autism-related deficits via dysregulated eIF4E-dependent translational control
Christos G. Gkogkas,Arkady Khoutorsky,Israeli Ran,Emmanouil Rampakakis,Tatiana Nevarko,Daniel B. Weatherill,Cristina Vasuta,Stephanie S. Yee,Morgan L. Truitt,Paul Dallaire,François Major,Paul Lasko,Davide Ruggero,Karim Nader,Jean-Claude Lacaille,Nahum Sonenberg +15 more
TL;DR: This article showed that deletion of the eukaryotic translation initiation factor 4E-binding protein 2 (4E-BP2) led to an increased ratio of excitatory to inhibitory synaptic inputs and autistic-like behaviors (that is, social interaction deficits, altered communication and repetitive/stereotyped behaviours).
Journal ArticleDOI
GLI1 is regulated through Smoothened-independent mechanisms in neoplastic pancreatic ducts and mediates PDAC cell survival and transformation
Olivier Nolan-Stevaux,Janet Lau,Morgan L. Truitt,Gerald C. Chu,Matthias Hebrok,Martin E. Fernández-Zapico,Douglas Hanahan +6 more
TL;DR: It is reported that multistage development of PDAC tumors is not affected by the deletion of Smo in the pancreas, demonstrating that autocrine Shh-Ptch-Smo signaling is not required in pancreatic ductal cells for PDAC progression.
