The CD47-signal regulatory protein alpha (SIRPa) interaction is a therapeutic target for human solid tumors
Stephen B. Willingham,Jens Peter Volkmer,Andrew J. Gentles,Debashis Sahoo,Piero Dalerba,Siddhartha Mitra,Jian Wang,Humberto Contreras-Trujillo,Robin Martin,Justin D. Cohen,Patricia Lovelace,Ferenc A. Scheeren,Mark P. Chao,Kipp Weiskopf,Chad Tang,Anne Kathrin Volkmer,Tejaswitha J. Naik,Theresa A. Storm,Adriane Mosley,Badreddin Edris,Seraina Schmid,Chris K. Sun,Mei-Sze Chua,Oihana Murillo,Pradeep S. Rajendran,Adriel C. Cha,Robert K. Chin,Dongkyoon Kim,Maddalena Adorno,Tal Raveh,Diane Tseng,Siddhartha Jaiswal,Per Øyvind Enger,Gary K. Steinberg,Gordon Li,Samuel So,Ravindra Majeti,Griffith R. Harsh,Matt De Van Rijn,Nelson N.H. Teng,John B. Sunwoo,Ash A. Alizadeh,Michael F. Clarke,Irving L. Weissman +43 more
TL;DR: All human solid tumor cells require CD47 expression to suppress phagocytic innate immune surveillance and elimination, showing that CD47 is a commonly expressed molecule on all cancers, its function to blockphagocytosis is known, and blockade of its function leads to tumor cell phagcytosis and elimination.
Journal ArticleDOI
Low surface expression of B7-1 (CD80) is an immunoescape mechanism of colon carcinoma
Iñigo Tirapu,Eduardo Huarte,Cristiana Guiducci,Ainhoa Arina,Mikel Zaratiegui,Oihana Murillo,Alvaro Gonzalez,Carmen Berasain,Pedro Berraondo,Puri Fortes,Jesús Prieto,Mario P. Colombo,Lieping Chen,Ignacio Melero +13 more
TL;DR: It is shown that unmutated CD80 is spontaneously expressed at low levels by mouse colon carcinoma cell lines and other transplantable tumor cell lines of various tissue origins, providing an explanation for the low levels of CD80 expression described in various human malignancies.
Journal ArticleDOI
Dendritic cells delivered inside human carcinomas are sequestered by interleukin-8.
Esperanza Feijoo,Carlos Alfaro,Guillermo Mazzolini,Patricia Serra,Iván Peñuelas,Ainhoa Arina,Eduardo Huarte,Iñigo Tirapu,Belen Palencia,Oihana Murillo,Juan Ruiz,Bruno Sangro,José A. Richter,Jesús Prieto,Ignacio Melero +14 more
TL;DR: It was found that tumors from patients suffering hepatocellular carcinoma, colorectal or pancreatic cancer were producing IL‐8 and that this chemokine attracted monocyte‐derived dendritic cells that uniformly express both IL•8 receptors CXCR1 and CX CR2, which blocked the chemotactic attraction of DCs by recombinant IL‐ 8.
Journal ArticleDOI
Multi-layered action mechanisms of CD137 (4-1BB)-targeted immunotherapies
TL;DR: As an agonist, anti-CD137 monoclonal antibody has entered Phase II clinical trials; elucidation of the mechanisms behind the antitumor efficacy requires further research in mice and patients to understand and rationally combine these new treatments.
Journal ArticleDOI
Long-term metabolic correction of Wilson's disease in a murine model by gene therapy.
Oihana Murillo,Daniel Moreno Luqui,Cristina Gazquez,Debora Martinez-Espartosa,Iñigo Navarro-Blasco,Jose Ignacio Monreal,Laura Guembe,Armando Moreno-Cermeño,Fernando J. Corrales,Jesús Prieto,Ruben Hernandez-Alcoceba,Gloria González-Aseguinolaza +11 more
TL;DR: It is demonstrated that AAV8-AAT-ATP7B-mediated gene therapy provides long-term correction of copper metabolism in a clinically relevant animal model of Wilson's disease providing support for future translational studies.
