Journal ArticleDOI
Head swivel on the ribosome facilitates translocation by means of intra-subunit tRNA hybrid sites
Andreas H. Ratje,Andreas H. Ratje,Justus Loerke,Aleksandra Mikolajka,Matthias Brünner,Peter W. Hildebrand,Agata L. Starosta,Alexandra Dönhöfer,Sean R. Connell,Paola Fucini,Thorsten Mielke,Paul C. Whitford,José N. Onuchic,Yanan Yu,Karissa Y. Sanbonmatsu,Roland K. Hartmann,Pawel A. Penczek,Daniel N. Wilson,Christian M. T. Spahn +18 more
TL;DR: Cryoelectron microscopy analysis is used to resolve two previously unseen subpopulations within Thermus thermophilus EF-G–ribosome complexes at subnanometre resolution and provides direct structural and mechanistic insight into the ‘missing link’ in terms of tRNA intermediates involved in the universally conserved translocation process.
Journal ArticleDOI
Translational regulation via L11: molecular switches on the ribosome turned on and off by thiostrepton and micrococcin.
Joerg Harms,Joerg Harms,Daniel N. Wilson,Daniel N. Wilson,Frank Schluenzen,Sean R. Connell,Sean R. Connell,Torsten Stachelhaus,Zaneta Zaborowska,Christian M. T. Spahn,Paola Fucini,Paola Fucini +11 more
TL;DR: The results suggest that L11 acts as a molecular switch to control L7 binding and plays a pivotal role in positioning one L7-CTD monomer on the G' subdomain ofEF-G to regulate EF-G turnover during protein synthesis.
Journal ArticleDOI
The oxazolidinone antibiotics perturb the ribosomal peptidyl-transferase center and effect tRNA positioning.
Daniel N. Wilson,Frank Schluenzen,Joerg Harms,Agata L. Starosta,Sean R. Connell,Paola Fucini +5 more
TL;DR: The crystal structure of the oxazolidinone linezolid bound to the Deinococcus radiodurans 50S ribosomal subunit is determined and a model whereby oxazolicinones impart their inhibitory effect by perturbing the correct positioning of tRNAs on the ribosome is presented.
Journal ArticleDOI
Detection and selective dissociation of intact ribosomes in a mass spectrometer
Adam A. Rostom,Paola Fucini,D R Benjamin,R Juenemann,Knud H. Nierhaus,Franz-Ulrich Hartl,Christopher M. Dobson,Carol V. Robinson +7 more
TL;DR: The results show that emerging mass spectrometric techniques can be used to characterize a fully functional biological assembly as well as its isolated components.
Journal ArticleDOI
Inhibition of peptide bond formation by pleuromutilins: the structure of the 50S ribosomal subunit from Deinococcus radiodurans in complex with tiamulin
TL;DR: The results show that tiamulin is located within the peptidyl transferase center (PTC) of the 50S ribosomal subunit with its tricyclic mutilin core positioned in a tight pocket at the A‐tRNA binding site, thus explaining the molecular mechanism of the antimicrobial activity of the pleuromutilin class of antibiotics.
