Journal ArticleDOI
Procedures for large-scale metabolic profiling of serum and plasma using gas chromatography and liquid chromatography coupled to mass spectrometry
Warwick B. Dunn,David Broadhurst,David Broadhurst,Paul Begley,Eva Zelena,Sue Francis-McIntyre,Nadine Anderson,Marie Brown,Joshau D Knowles,Antony Halsall,John N. Haselden,Andrew W. Nicholls,Ian D. Wilson,Douglas B. Kell,Royston Goodacre +14 more
TL;DR: The experimental workflow for long-term and large-scale metabolomic studies involving thousands of human samples with data acquired for multiple analytical batches over many months and years is described.
Journal ArticleDOI
Development of a robust and repeatable UPLC-MS method for the long-term metabolomic study of human serum.
Eva Zelena,Warwick B. Dunn,David Broadhurst,Sue Francis-McIntyre,Kathleen M. Carroll,Paul Begley,Steve O'Hagan,Joshua Knowles,Antony Halsall,Ian D. Wilson,Douglas B. Kell +10 more
TL;DR: The observed drift in instrumental performance over time and its improvement with adjustment of the length of analytical block are described and allowed the authors to prepare SOPs for "fit for purpose" long-term UPLC-MS metabolomic studies, such as are being employed in the HUSERMET project.
Journal ArticleDOI
Mass spectrometry tools and metabolite-specific databases for molecular identification in metabolomics
Marie Brown,Warwick B. Dunn,Paul D. Dobson,Yogendra Patel,Catherine L. Winder,Sue Francis-McIntyre,Paul Begley,Kathleen M. Carroll,David Broadhurst,Andy Tseng,Neil Swainston,Irena Spasic,Royston Goodacre,Douglas B. Kell +13 more
TL;DR: The combination of accurate mass data for a large collection of metabolites, theoretical isotope abundance data and knowledge of the different ion types detected provided a greater number of electrospray mass spectrometric signals which were putatively identified and with greater confidence in the samples studied.
Journal ArticleDOI
Molecular phenotyping of a UK population: defining the human serum metabolome.
Warwick B. Dunn,Wanchang Lin,Wanchang Lin,David Broadhurst,David Broadhurst,Paul Begley,Paul Begley,Marie Brown,Marie Brown,Eva Zelena,Andrew A. Vaughan,Antony Halsall,Nadine Harding,Joshua Knowles,Sue Francis-McIntyre,Andy Tseng,David I. Ellis,Steve O'Hagan,Gill Aarons,Boben Benjamin,Stephen Chew-Graham,Carly Moseley,Paula Potter,Catherine L. Winder,Catherine Potts,Paula Thornton,Catriona McWhirter,Mohammed Zubair,Martin Pan,Alistair Burns,J. Kennedy Cruickshank,J. Kennedy Cruickshank,Gordon C Jayson,Nitin Purandare,Frederick C. W. Wu,J. D. Finn,John N. Haselden,Andrew W. Nicholls,Ian D. Wilson,Ian D. Wilson,Royston Goodacre,Douglas B. Kell +41 more
TL;DR: The Husermet study as discussed by the authors used a large scale and non-targeted chromatographic MS-based metabolomics study, using samples from over 1,000 individuals, to provide a comprehensive measurement of their serum metabolomes.
Journal ArticleDOI
Development and Performance of a Gas Chromatography−Time-of-Flight Mass Spectrometry Analysis for Large-Scale Nontargeted Metabolomic Studies of Human Serum
Paul Begley,Sue Francis-McIntyre,Warwick B. Dunn,David Broadhurst,Antony Halsall,Andy Tseng,Joshua Knowles,Royston Goodacre,Douglas B. Kell +8 more
TL;DR: A method for the preparation and GC-TOF-MS analysis of human serum samples has been developed and evaluated for application in long-term metabolomic studies and showed the ability to define metabolite differences between samples from a population and samples spiked with metabolites standards.
