Journal ArticleDOI
Role of acidic pH in the susceptibility of intraphagocytic methicillin-resistant Staphylococcus aureus strains to meropenem and cloxacillin.
Sandrine Lemaire,Françoise Van Bambeke,Marie-Paule Mingeot-Leclercq,Youri Glupczynski,Paul M. Tulkens +4 more
TL;DR: The restoration of susceptibility of intraphagocytic of MRSA to MEM and CLX is due to the acidic pH prevailing in phagolysosomes and is mediated by an enhanced binding to penicillin-binding proteins.
Journal ArticleDOI
Activity of moxifloxacin against intracellular community-acquired methicillin-resistant Staphylococcus aureus: comparison with clindamycin, linezolid and co-trimoxazole and attempt at defining an intracellular susceptibility breakpoint
Sandrine Lemaire,Klaudia Kosowska-Shick,Peter C. Appelbaum,Youri Glupczynski,Françoise Van Bambeke,Paul M. Tulkens +5 more
TL;DR: Moxifloxacin is active against both extracellular and intracellular CA-MRSA if the MIC is low, and is more effective than clindamycin, co-trimoxazole and linezolid.
Journal ArticleDOI
Cellular accumulation of fluoroquinolones is not predictive of their intracellular activity: studies with gemifloxacin, moxifloxacin and ciprofloxacin in a pharmacokinetic/pharmacodynamic model of uninfected and infected macrophages
Coralie M. Vallet,Béatrice Marquez,Eva Ngabirano,Sandrine Lemaire,Marie Paula Mingeot-Leclercq,Paul M. Tulkens,Françoise Van Bambeke +6 more
TL;DR: Data suggest that cellular accumulation of fluoroquinolones is largely a self-defeating process as far as activity is concerned, with the intracellular drug made inactive in proportion to its accumulation level.
Journal ArticleDOI
Cellular toxicity of gentamicin.
Jacques Libert,Paul E. Ketelbant-Balasse,François Van Hoof,Genevieve G. Aubert-Tulkens,Paul M. Tulkens +4 more
TL;DR: Subconjunctival injections of gentamicin induced a lysosomal storage process within the conjunctival fibroblasts in rats, rabbits, and humans, and it is believed human kidneys might present similar alterations.
Journal ArticleDOI
Profile of a Novel Anionic Fluoroquinolone-Delafloxacin.
TL;DR: Delafloxacin shows favorable pharmacological properties, suitability for intravenous, oral, or switch dosing, the lack of many clinically significant drug-drug interactions, and acceptable adverse event profile in registration clinical trials supports its use in the treatment of acute bacterial skin and skin structure infections, and potentially in other infections, where resistance to other agents, safety, and/or the need for early discharge is of concern.
