Journal ArticleDOI
Sampling times in micronucleus testing.
TL;DR: It is evident that for screening purposes two sampling times at 24 and 48 h are sufficient to detect clastogens as well as aneugens and no qualitative differences were observed between the two sexes.
Journal ArticleDOI
Optimisation of the cell cultivation methods in the embryonic stem cell test results in an increased differentiation potential of the cells into strong beating myocard cells.
Ann De Smedt,Margino Steemans,Marlies De Boeck,Annelieke K. Peters,Bas-jan van der Leede,Freddy Van Goethem,Ann Lampo,Philippe Vanparys +7 more
TL;DR: A stringent control of the cell growth during subcultivation and a standardised hanging drop culture method and a non-enzymatic cell harvest resulted in a decreased variability in the size of embryonic bodies, an increase in the number of acceptable tests and a significant increase of the differentiation potential of embryonic cells into strong beating myocardium, which made scoring less time consuming.
Journal ArticleDOI
Interlaboratory validation of a CD71-based flow cytometric method (Microflow) for the scoring of micronucleated reticulocytes in mouse peripheral blood.
Dorothea K. Torous,Nikki E. Hall,Anne H. Illi-Love,Marilyn S. Diehl,Karin Cederbrant,Kerstin Sandelin,Ingrid Pontén,George Bolcsfoldi,Lynnette R. Ferguson,Amira Pearson,Jenness B. Majeska,James P. Tarca,Geoffrey M. Hynes,Anthony M. Lynch,James P. McNamee,P. V. Bellier,Monique Parenteau,David H. Blakey,Janet Bayley,Bas-jan van der Leede,Philippe Vanparys,Philip R. Harbach,Shuou Zhao,Anthony L. Filipunas,Carol W. Johnson,Carol R. Tometsko,Stephen D. Dertinger +26 more
TL;DR: The overall performance of mouse peripheral blood micronucleus tests is enhanced by the use of the flow cytometric scoring procedure, which offered advantages relative to microscopy‐based scoring, including a greater number of cells analyzed, much faster analysis times, and a greater degree of objectivity.
Journal ArticleDOI
ECVAM prevalidation of three cell transformation assays.
Philippe Vanparys,Raffaella Corvi,Marilyn Aardema,Laura Gribaldo,Makoto Hayashi,Sebastian Hoffmann,Leonard M. Schechtman +6 more
TL;DR: The Validation Management Team concluded that standardised protocols are now available that should be the basis for future use and are reproducible within- and between-laboratories.
Journal ArticleDOI
Automated analysis of contractility in the embryonic stem cell test, a novel approach to assess embryotoxicity.
Annelieke K. Peters,Gert Van de Wouwer,Barbara Weyn,Geert R. Verheyen,Philippe Vanparys,Jacques Van Gompel +5 more
TL;DR: The automated image recording of contractile cardiomyocyte-like cells in the EST allows for an unbiased high throughput method to assess the embryotoxic potency of test compounds, resulting in an outcome comparable to manual analysis.
