Journal ArticleDOI
College of American pathologists' laboratory standards for next-generation sequencing clinical tests
Nazneen Aziz,Qin Zhao,Lynn Bry,Denise K. Driscoll,Birgit Funke,Jane Gibson,Wayne W. Grody,Madhuri Hegde,Gerald A. Hoeltge,Debra G.B. Leonard,Jason D. Merker,Rakesh Nagarajan,Linda A. Palicki,Ryan S. Robetorye,Iris Schrijver,Karen E. Weck,Karl V. Voelkerding +16 more
TL;DR: This report describes the important issues considered by the CAP committee during the development of the new checklist requirements, which address documentation, validation, quality assurance, confirmatory testing, exception logs, monitoring of upgrades, variant interpretation and reporting, incidental findings, data storage, version traceability and data transfer confidentiality.
Journal ArticleDOI
A Model Study of In Silico Proficiency Testing for Clinical Next-Generation Sequencing
Eric J. Duncavage,Haley J. Abel,Jason D. Merker,John B Bodner,Qin Zhao,Karl V. Voelkerding,John D. Pfeifer +6 more
TL;DR: The results indicate that in silico proficiency testing is a feasible approach for methods-based proficiency testing, and demonstrate that the sensitivity and specificity of current next-generation sequencing bioinformatics across clinical laboratories are high.
Journal ArticleDOI
Genetic and Functional Identifying of Novel STAT1 Loss-of-Function Mutations in Patients with Diverse Clinical Phenotypes
Xuemei Chen,Junjie Chen,R. Chen,Hui Mou,Gan Sun,Lu Yang,Yan-Jun Jia,Qin Zhao,Wen-Juan Wen,Lina Zhou,Yuan Ding,Xuemei Tang,Jun Yang,Yunfei An,Xiaodong Zhao +14 more
TL;DR: The susceptibility to infections and immunological deficiency phenotypes, observed in AD-LOF patients, confirms the importance of STAT1 in host-pathogen interaction and immunity, but variability in the nature and extent of these phenotypes suggests that functional analysis is required to identify accurately novel, heterozygous STAT1 mutations, associated with pathogenicity.
Journal ArticleDOI
A novel homozygous RAG1 mutation in a girl presenting with granulomas and alopecia capitis totalis
TL;DR: These patients present with generalized granulomas, severe complications after viral hypogammaglobulinemia, and various autoim-mune manifestations.
Journal ArticleDOI
WASp Deficiency Selectively Affects the TCR Diversity of Different Memory T Cell Subsets in WAS Chimeric Mice
Wenyan Li,Yan-Jun Jia,Yanping Wang,Qin Zhao,Lu Yang,Ting Zeng,Linlin Niu,Rongxin Dai,Yanan Li,Xiaodong Zhao,Junfeng Wu +10 more
TL;DR: WASp deficiency selectively affected the T CR diversity of different memory T cell subsets, and it had more impact on the TCRVβ diversity of CD4+ TEM cells than CD8+ TCM cells.
