Exoelectrogenic bacteria that power microbial fuel cells

While electrochemical characterization of enzymes immobilized on electrodes has become common, there is still a need for reliable quantitative methods for study of electron transfer between living cells and conductive surfaces. This work describes growth of thin (<20 microm) Geobacter sulfurreducens biofilms on polished glassy carbon electrodes, using stirred three-electrode anaerobic bioreactors controlled by potentiostats and nondestructive voltammetry techniques for characterization of viable biofilms. Routine in vivo analysis of electron transfer between bacterial cells and electrodes was performed, providing insight into the main redox-active species participating in electron transfer to electrodes. At low scan rates, cyclic voltammetry revealed catalytic electron transfer between cells and the electrode, similar to what has been observed for pure enzymes attached to electrodes under continuous turnover conditions. Differential pulse voltammetry and electrochemical impedance spectroscopy also revealed features that were consistent with electron transfer being mediated by an adsorbed catalyst. Multiple redox-active species were detected, revealing complexity at the outer surfaces of this bacterium. These techniques provide the basis for cataloging quantifiable, defined electron transfer phenotypes as a function of potential, electrode material, growth phase, and culture conditions and provide a framework for comparisons with other species or communities.

Microbial Biofilm Voltammetry: Direct Electrochemical Characterization of Catalytic Electrode-Attached Biofilms

Microbial fuel cells (MFCs) are devices that can use bacterial metabolism to produce an electrical current from a wide range organic substrates. Due to the promise of sustainable energy production from organic wastes, research has intensified in this field in the last few years. While holding great promise only a few marine sediment MFCs have been used practically, providing current for low power devices. To further improve MFC technology an understanding of the limitations and microbiology of these systems is required. Some researchers are uncovering that the greatest value of MFC technology may not be the production of electricity but the ability of electrode associated microbes to degrade wastes and toxic chemicals. We conclude that for further development of MFC applications, a greater focus on understanding the microbial processes in MFC systems is required.

Sustainable wastewater treatment: How might microbial fuel cells contribute

Microbial fuel cells (MFCs) have attracted considerable interest due to their potential in renewable electrical power generation using the broad diversity of biomass and organic substrates. However, the difficulties in achieving high power densities and commercially affordable electrode materials have limited their industrial applications to date. Carbon materials, which can exhibit a wide range of different morphologies and structures, usually possess physiological activity to interact with microorganisms and are therefore fast-emerging electrode materials. As the anode, carbon materials can significantly promote interfacial microbial colonization and accelerate the formation of extracellular biofilms, which eventually promotes the electrical power density by providing a conductive microenvironment for extracellular electron transfer. As the cathode, carbon-based materials can function as catalysts for the oxygen-reduction reaction, showing satisfying activities and efficiencies nowadays even reaching the performance of Pt catalysts. Here, first, recent advancements on the design of carbon materials for anodes in MFCs are summarized, and the influence of structure and surface functionalization of different types of carbon materials on microorganism immobilization and electrochemical performance is elucidated. Then, synthetic strategies and structures of typical carbon-based cathodes in MFCs are briefly presented. Furthermore, future applications of carbon-electrode-based MFC devices in the energy, environmental, and biological fields are discussed, and the emerging challenges in transferring them from laboratory to industrial scale are described.

In the practice of medicine, many fundamental biological pathways that require tight on/off control, such as inflammation and circulatory homeostasis, are regulated by serine proteinases, but we rarely consider the unique protease inhibitors that, in turn, regulate these proteases. The serpins are a family of proteins with a shared tertiary structure, whose members largely act as serine protease inhibitors, found in all forms of life, ranging from viruses, bacteria, and archaea to plants and animals. These proteins represent up to 2–10% of proteins in the human blood and are the third most common protein family.
