PD-1/PD-L1 binding studies using microscale thermophoresis.
Romain Magnez,Bryan Thiroux,Bryan Thiroux,Solenne Taront,Solenne Taront,Zacharie Segaoula,Zacharie Segaoula,Bruno Quesnel,Bruno Quesnel,Xavier Thuru,Xavier Thuru +10 more
TL;DR: A protocol using microscale thermophoresis to determine the binding affinity of the PD-1/PD-L1 couple, which is involved in tumour escape processes, without purification of the target protein from cell lysates is described.
Journal ArticleDOI
Toward the Discovery of a Novel Class of YAP–TEAD Interaction Inhibitors by Virtual Screening Approach Targeting YAP–TEAD Protein–Protein Interface
Floriane Gibault,Mathilde Coevoet,Manon Sturbaut,Amaury Farce,Nicolas Renault,Frédéric Allemand,Jean-François Guichou,Anne-Sophie Drucbert,Catherine Foulon,Romain Magnez,Xavier Thuru,Matthieu Corvaisier,Guillemette Huet,Philippe Chavatte,Patricia Melnyk,Fabrice Bailly,Philippe Cotelle +16 more
TL;DR: A first promising hit with micromolar inhibition in the luciferase gene reporter assay was discovered, and this hit also decreased mRNA levels of TEAD target genes.
Journal ArticleDOI
Discovery of a cryptic site at the interface 2 of TEAD - Towards a new family of YAP/TAZ-TEAD inhibitors.
Manon Sturbaut,Fabrice Bailly,Mathilde Coevoet,Pasquale Sileo,Martine Pugnière,Maxime Liberelle,Romain Magnez,Xavier Thuru,Marie-Christine Chartier-Harlin,Patricia Melnyk,Muriel Gelin,Frédéric Allemand,Jean-François Guichou,Philippe Cotelle +13 more
TL;DR: In this paper, a set of trisubstituted pyrazoles which bind to hTEAD2 at the interface 2 revealing for the first time a cryptic pocket created by the movement of the phenol ring of Y382.
Journal ArticleDOI
IL-2 is inactivated by the acidic pH environment of tumors enabling engineering of a pH-selective mutein.
Silvia Gaggero,Jonathan Martínez-Fábregas,A. Cozzani,Paul K. Fyfe,Malo Leprohon,F. Emil Thomasen,Hauke Winkelmann,Romain Magnez,Alberto G Conti,Stephan Wilmes,Elizabeth Pohler,Manuel van Gijsel Bonnello,Xavier Thuru,Bruno Quesnel,Fabrice Soncin,Jacob Piehler,Kresten Lindorff-Larsen,Rahul Roychoudhuri,Ignacio Moraga,Suman Mitra +19 more
TL;DR: In this paper , the authors show that the activity of interleukin-2 (IL-2), a cytokine critical to T cell immunity, is profoundly affected by pH, limiting IL-2 signaling within the acidic environment of tumors.
Journal ArticleDOI
MUC4-ErbB2 Oncogenic Complex: Binding studies using Microscale Thermophoresis.
Maxime Liberelle,Romain Magnez,Xavier Thuru,Yamina Bencheikh,Séverine Ravez,Camille Quenon,Anne-Sophie Drucbert,Catherine Foulon,Patricia Melnyk,Isabelle Van Seuningen,Nicolas Lebegue +10 more
TL;DR: New information is provided regarding the M UC4-ErbB2 interaction at the biophysical level and the presence of the three EGF domains of MUC4 is sufficient to provide efficient interaction, which will be of high interest for the other known membrane mucins forming oncogenic complexes with ErbBs at the cancer cell surface.
