Mismatch repair deficiency predicts response of solid tumors to PD-1 blockade
Dung T. Le,Dung T. Le,Jennifer N. Durham,Jennifer N. Durham,Kellie N. Smith,Hao Wang,Bjarne Bartlett,Bjarne Bartlett,Laveet K. Aulakh,Laveet K. Aulakh,Steve Lu,Steve Lu,Holly Kemberling,Cara Wilt,Brandon Luber,Fay Wong,Fay Wong,Nilofer S. Azad,Agnieszka A. Rucki,Daniel A. Laheru,Ross C. Donehower,Atif Zaheer,George A. Fisher,Todd S. Crocenzi,James J. Lee,Tim F. Greten,Austin G. Duffy,Kristen K. Ciombor,Aleksandra Eyring,Bao H. Lam,Andrew K. Joe,S. Peter Kang,Matthias Holdhoff,Ludmila Danilova,Leslie Cope,Christian F. Diaz +57 more
TL;DR: Evaluating the efficacy of PD-1 blockade in patients with advanced mismatch repair–deficient cancers across 12 different tumor types showed that colorectal cancers with mismatch repair deficiency were sensitive to immune checkpoint blockade with antibodies to programmed death receptor–1 (PD-1).
Journal ArticleDOI
Glucose deprivation contributes to the development of KRAS pathway mutations in tumor cells.
Jihye Yun,Carlo Rago,Ian Cheong,Ray Pagliarini,Ray Pagliarini,Philipp Angenendt,Harith Rajagopalan,Harith Rajagopalan,Kerstin Schmidt,James K V Willson,Sandy D. Markowitz,Shibin Zhou,Luis A. Diaz,Victor E. Velculescu,Christoph Lengauer,Christoph Lengauer,Kenneth W. Kinzler,Bert Vogelstein,Nickolas Papadopoulos +18 more
TL;DR: Studying the transcriptomes of paired colorectal cancer cell lines that differed only in the mutational status of their KRAS or BRAF genes, it is suggested that glucose deprivation can drive the acquisition of KRAS pathway mutations in human tumors.
Journal ArticleDOI
Inactivating mutations of the chromatin remodeling gene ARID2 in hepatocellular carcinoma
Meng Li,Hong Zhao,Xiaosong Zhang,Laura D. Wood,Robert A. Anders,Michael A. Choti,Timothy M. Pawlik,Hubert Darius J. Daniel,Rajesh Kannangai,G. Johan A. Offerhaus,Victor E. Velculescu,Linfang Wang,Shibin Zhou,Bert Vogelstein,Ralph H. Hruban,Nick Papadopoulos,Jianqiang Cai,Michael Torbenson,Kenneth W. Kinzler +18 more
TL;DR: Exomic sequencing of ten hepatitis C virus-associated hepatocellular carcinomas and subsequent evaluation of additional affected individuals discovered novel inactivating mutations of ARID2 in four major subtypes of HCC, suggesting that ARIDs is a tumor suppressor gene that is relatively commonly mutated in this tumor subtype.
Journal ArticleDOI
Transcriptional programs of neoantigen-specific TIL in anti-PD-1-treated lung cancers.

Forde,Srinivasan Yegnasubramanian,Hongkai Ji,Drew M. Pardoll,Kellie N. Smith +62 more
TL;DR: In this paper, single-cell RNA sequencing and T cell receptor sequencing are combined to identify transcriptional programs specific to mutation-associated neoantigen-specific T cells in non-small cell lung cancers treated with anti-PD-1, providing insights into resistance to PD-1 blockade.
Journal ArticleDOI
Applications of liquid biopsies for cancer.
Austin Mattox,Chetan Bettegowda,Shibin Zhou,Nickolas Papadopoulos,Kenneth W. Kinzler,Bert Vogelstein +5 more
TL;DR: This data indicates that liquid biopsies have the potential to detect, characterize, and monitor cancers earlier than is possible with conventional approaches.
