The sensing of poorly deformable red blood cells by the human spleen can be mimicked in vitro
Guillaume Deplaine,Guillaume Deplaine,Guillaume Deplaine,Innocent Safeukui,Innocent Safeukui,Fakhri Jeddi,François Lacoste,Valentine Brousse,Sylvie Perrot,Sylvie Perrot,Sylvestre Biligui,Micheline Guillotte,Micheline Guillotte,Corinne Guitton,Safi Dokmak,Béatrice Aussilhou,Alain Sauvanet,Dominique Cazals Hatem,François Paye,Marc Thellier,Dominique Mazier,Geneviève Milon,Narla Mohandas,Odile Mercereau-Puijalon,Odile Mercereau-Puijalon,Peter H. David,Peter H. David,Pierre Buffet +27 more
TL;DR: In vitro results directly confirm the importance of the mechanical sensing of RBCs by the human spleen and designed a sorting device where a mixture of 5- to 25-μm-diameter microbeads mimics the geometry of narrow and short interendothelial splenic slits.
Journal ArticleDOI
Role of the integument in insect immunity: epicuticular abrasion and induction of cecropin synthesis in cuticular epithelial cells.
Paul T. Brey,Won-Jae Lee,Minoru Yamakawa,Yoshiko Koizumi,Sylvie Perrot,Madeleine Francois,Masaaki Ashida +6 more
TL;DR: The data illustrate that the ectodermally derived lepidopteran larval integument, when challenged by live bacteria or their cell wall components, mounts an immune response, and the insect exoskeleton is indeed actively participating in defense.
Journal ArticleDOI
A switch in infected erythrocyte deformability at the maturation and blood circulation of Plasmodium falciparum transmission stages
Marta Tibúrcio,Makhtar Niang,Guillaume Deplaine,Sylvie Perrot,Emmanuel Bischoff,Papa Alioune Ndour,Francesco Silvestrini,Ayman Khattab,Geneviève Milon,Peter H. David,Max R. Hardeman,Kenneth D. Vernick,Robert W. Sauerwein,Peter R. Preiser,Odile Mercereau-Puijalon,Pierre Buffet,Pietro Alano,Catherine Lavazec,Catherine Lavazec +18 more
TL;DR: It is shown that mature GIEs are more deformable than immature stages using ektacytometry and microsphiltration methods, and that a switch in cellular deformability in the transition from immature to mature gametocytes is accompanied by the deassociation of parasite-derived STEVOR proteins from the infected erythrocyte membrane.
Journal ArticleDOI
Quantitative assessment of sensing and sequestration of spherocytic erythrocytes by the human spleen
Innocent Safeukui,Innocent Safeukui,Pierre Buffet,Guillaume Deplaine,Guillaume Deplaine,Guillaume Deplaine,Sylvie Perrot,Sylvie Perrot,Valentine Brousse,Alioune Ndour,Marie Nguyen,Odile Mercereau-Puijalon,Odile Mercereau-Puijalon,Peter H. David,Peter H. David,Geneviève Milon,Narla Mohandas +16 more
TL;DR: The results of the present study define the critical extent of surface area loss leading to splenic entrapment and identify an adaptive volume regulation mechanism that allows spherocytic RBCs to prolong their life span in circulation.
Journal ArticleDOI
Proteomic Analysis of Intact Flagella of Procyclic Trypanosoma brucei Cells Identifies Novel Flagellar Proteins with Unique Sub-localization and Dynamics
Ines Subota,Daria Julkowska,Laetitia Vincensini,Nele Reeg,Johanna Buisson,Thierry Blisnick,Diego Huet,Sylvie Perrot,Julien Santi-Rocca,Magalie Duchateau,Véronique Hourdel,Jean-Claude Rousselle,Nadège Cayet,Abdelkader Namane,Julia Chamot-Rooke,Philippe Bastin +15 more
TL;DR: The purification of intact flagella from the procyclic stage of Trypanosoma brucei using mechanical shearing demonstrated very different dynamics from one protein to the other, but also according to the stage of construction and the age of the flagellum.
