Journal ArticleDOI
The hypoxic tumour microenvironment.
TL;DR: The most relevant findings describing the influence of hypoxia and the contribution of HIF activation on the major components of the tumour microenvironment are reviewed, and their role in cancer development and progression is summarised.
Journal ArticleDOI
p53 mutants cooperate with HIF-1 in transcriptional regulation of extracellular matrix components to promote tumor progression.
Ivano Amelio,Mara Mancini,Varvara Petrova,Rob A. Cairns,Polina Vikhreva,Sara Nicolai,Alberto Marini,Alexey A Antonov,John Le Quesne,John Le Quesne,Juvenal Dario Baena Acevedo,Kate Dudek,Gabriella Sozzi,Ugo Pastorino,Richard A. Knight,Tak W. Mak,Gerry Melino,Gerry Melino +17 more
TL;DR: It is demonstrated that in non-small cell lung cancer (NSCLC), p53 mutants exert a gain-of-function (GOF) effect on HIF-1, thus regulating a selective gene expression signature involved in protumorigenic functions.
Journal ArticleDOI
Discovery of Novel Isatin-Based p53 Inducers.
Pavel B. Davidovich,Vasilisa Aksenova,Varvara Petrova,D. Tentler,Orlova Dd,S. N. Smirnov,Vladislav V. Gurzhiy,Andrei L. Okorokov,A. V. Garabadzhiu,Gerry Melino,Nickolai A. Barlev,Vyacheslav G. Tribulovich +11 more
TL;DR: The hit compounds activated and stabilized p53, as shown by Western blotting, at higher rates than the well-known positive control Nutlin-3.
Journal ArticleDOI
TAp73 upregulates IL-1β in cancer cells: Potential biomarker in lung and breast cancer?
P. N. Vikhreva,Varvara Petrova,Tarik Gokbulut,Ilias Pestlikis,Mara Mancini,Nicola Di Daniele,Richard A. Knight,Gerry Melino,Ivano Amelio +8 more
TL;DR: Results show that TAp73β upregulates pro-IL-1β mRNA and processed IL-1 β protein, and analysis of breast and lung cancer patient cohorts demonstrated that interaction between p73 and IL- 1β predicts a negative survival outcome in these human cancers.
Journal ArticleDOI
Novel isatin-derived molecules activate p53 via interference with Mdm2 to promote apoptosis.
Olga A. Fedorova,Alexandra Daks,Varvara Petrova,Alexey Petukhov,Larissa Lezina,Oleg Shuvalov,Pavel B. Davidovich,Darya Kriger,Ekaterina Lomert,Dmitry Tentler,Victor Kartsev,Burhan Uyanik,Vyacheslav G. Tribulovich,Oleg N. Demidov,Gerry Melino,Nickolai A. Barlev +15 more
TL;DR: These novel pharmacological activators of p53 can serve as valuable molecular tools for probing p53-positive tumors and set up the stage for development of new anti-cancer drugs.
