Toll-Like Receptors Induce Signal-Specific Reprogramming of the Macrophage Lipidome
Wei Yuan Hsieh,Quan D. Zhou,Autumn G. York,Kevin J. Williams,Philip O. Scumpia,Eliza B. Kronenberger,Xen Ping Hoi,Baolong Su,Xun Chi,Viet L. Bui,Elvira Khialeeva,Amber Kaplan,Young Min Son,Ajit S. Divakaruni,Jie Sun,Stephen T. Smale,Richard A. Flavell,Steven J. Bensinger +17 more
TL;DR: In this paper, the authors used complementary "shotgun" and isotope tracer mass spectrometry approaches to define the changes in lipid biosynthesis, import, and composition of macrophages induced by various Toll-like receptors (TLRs) and inflammatory cytokines.
Journal ArticleDOI
Interferon-mediated reprogramming of membrane cholesterol to evade bacterial toxins
Quan D. Zhou,Quan D. Zhou,Xun Chi,Min Sub Lee,Wei Yuan Hsieh,Jonathan J. Mkrtchyan,An-Chieh Feng,Cuiwen He,Autumn G. York,Autumn G. York,Autumn G. York,Viet L. Bui,Eliza B. Kronenberger,Alessandra Ferrari,Xu Xiao,Allison E. Daly,Elizabeth J. Tarling,Robert Damoiseaux,Philip O. Scumpia,Stephen T. Smale,Kevin J. Williams,Peter Tontonoz,Steven J. Bensinger +22 more
TL;DR: It is reported that interferon (IFN) signals convey resistance to CDC-induced pores on macrophages and neutrophils, and interferons promote host cell resistance to bacterial cytolysins by decreasing cholesterol synthesis and promoting the esterification of cholesterol, which alters the availability of this pool of ‘free’ cholesterol needed for pore formation.
Journal ArticleDOI
Development and Application of FASA, a Model for Quantifying Fatty Acid Metabolism Using Stable Isotope Labeling
Joseph P. Argus,Moses Q. Wilks,Quan D. Zhou,Quan D. Zhou,Wei Yuan Hsieh,Elvira Khialeeva,Xen Ping Hoi,Viet L. Bui,Shili Xu,Amy K. Yu,Eric Wang,Harvey R. Herschman,Kevin J. Williams,Steven J. Bensinger +13 more
TL;DR: Application of FASA demonstrated that elongation can be a major contributor to cellular fatty acid content and showed that distinct pro-inflammatory stimuli specifically reprogram homeostasis of fatty acids by differential utilization of synthetic and elongation pathways in macrophages.
Journal ArticleDOI
Generalized Cytokine Increase in the Setting of a Multisystem Clinical Disorder and Carcinoid Syndrome Associated with a Novel NLRP12 Variant
Noam Jacob,Sonya Dasharathy,Viet L. Bui,Jihane N. Benhammou,Wayne W. Grody,Ram Raj Singh,Joseph R. Pisegna +6 more
TL;DR: A novel variant in the NLRP12 gene is identified in a 63-year-old woman and her daughter, who presented with an unusual clinical syndrome that differs from autoinflammatory disorders previously reported in association with theNLRP subfamily gene mutations.
