Journal ArticleDOI
Linkage disequilibrium-dependent architecture of human complex traits shows action of negative selection.
Steven Gazal,Hilary K. Finucane,Hilary K. Finucane,Hilary K. Finucane,Nicholas A. Furlotte,Po-Ru Loh,Po-Ru Loh,Pier Francesco Palamara,Pier Francesco Palamara,Xuanyao Liu,Xuanyao Liu,Armin P. Schoech,Armin P. Schoech,Brendan Bulik-Sullivan,Benjamin M. Neale,Benjamin M. Neale,Alexander Gusev,Alexander Gusev,Alkes L. Price,Alkes L. Price +19 more
TL;DR: It is determined that SNPs with low LLD have significantly larger per-SNP heritability and that roughly half of this effect can be explained by functional annotations negatively correlated with LLD, such as DNase I hypersensitivity sites (DHSs).
Journal ArticleDOI
Trans Effects on Gene Expression Can Drive Omnigenic Inheritance.
TL;DR: This work provides a formal model in which genetic contributions to complex traits are partitioned into direct effects from core genes and indirect effects from peripheral genes acting in trans, proposing that most heritability is driven by weak trans-eQTL SNPs, whose effects are mediated through peripheral genes to impact the expression of core genes.
Journal ArticleDOI
Deep whole-genome sequencing of 100 southeast Asian Malays.
Lai-Ping Wong,Rick Twee-Hee Ong,Wan-Ting Poh,Xuanyao Liu,Peng Chen,Ruoying Li,Kevin Lam,Nisha Esakimuthu Pillai,Kar Seng Sim,Haiyan Xu,Ngak-Leng Sim,Shu Mei Teo,Jia Nee Foo,Linda Wei-Lin Tan,Yenly Lim,Seok-Hwee Koo,Linda Seo-Hwee Gan,Ching-Yu Cheng,Ching-Yu Cheng,Sharon Wee,Eric P.H. Yap,Pauline Crystal Ng,Wei-Yen Lim,Richie Soong,Markus R. Wenk,Tin Aung,Tin Aung,Tien Yin Wong,Tien Yin Wong,Chiea Chuen Khor,Peter Little,Kee Seng Chia,Yik Ying Teo +32 more
TL;DR: A comparison of the fidelity of genotype imputation of Malays indicated that a population-specific reference panel, such as the SSMP, outperforms a cosmopolitan panel with larger number of individuals for common SNPs and the higher sensitivity at detecting low-frequency and rare variants.
Journal ArticleDOI
Leveraging molecular quantitative trait loci to understand the genetic architecture of diseases and complex traits.
Farhad Hormozdiari,Farhad Hormozdiari,Steven Gazal,Steven Gazal,Bryce van de Geijn,Bryce van de Geijn,Hilary K. Finucane,Hilary K. Finucane,Chelsea J.-T. Ju,Po-Ru Loh,Po-Ru Loh,Armin P. TL;DR: A new set of functional annotations based on fine-mapped molecular quantitative trait loci from GTEx and BLUEPRINT consortium data are enriched for disease heritability across 41 diseases and complex traits.

Trans effects on gene expression can drive omnigenic inheritance
TL;DR: This work provides a formal model in which genetic contributions to complex traits can be partitioned into direct effects from core genes, and indirect effects from peripheral genes acting as trans-regulators, and proposes a framework for understanding key features of the architecture of complex traits.
